Decoding the role of macrophages in periodontitis and type 2 diabetes using single-cell RNA-sequencing

Panagiota Agrafioti et al.

Summary: By using single-cell RNA-sequencing, this study revealed the heterogeneity and transcriptional heterogeneity of macrophages in periodontitis. The study found that macrophages in periodontitis express both pro-inflammatory and anti-inflammatory markers, and their polarization is closely associated with the inflammatory response.

MG53 represses high glucose-induced inflammation and angiogenesis in human retinal endothelial cells by repressing the EGR1/STAT3 axis

Kun-Ming Cui et al.

Summary: In this study, the researchers investigated the role and mechanisms of MG53 in ameliorating the dysfunction induced by high glucose in human retinal microvascular endothelial cells (HRECs). They found that MG53 can improve cell damage caused by high glucose by repressing the EGR1/STAT3 signaling pathway and may have therapeutic potential in diabetic retinopathy.

Association of enhanced circulating trimethylamine N-oxide with vascular endothelial dysfunction in periodontitis patients

Jiamin Zhou et al.

Summary: This study demonstrates for the first time that circulating TMAO levels are increased in patients with Stage III-IV periodontitis, and correlated with vascular endothelial dysfunction. These findings may provide a novel insight into the mechanism of vascular endothelial dysfunction in patient with periodontitis via TMAO-downregulated EPC functions.

Neuropeptides as the Shared Genetic Crosstalks Linking Periodontitis and Major Depression Disorder

Changsheng Sun et al.

Summary: This study explored the association between major depressive disorder (MDD) and periodontitis using crosstalk genes and neuropeptides. Neuropeptide genes ADM, IGF2, PDYN, and RETN were found to intersect between periodontitis and MDD, with the crosstalk gene FOSB potentially playing a significant role in the disease mechanisms. These results provide a basis for further research and validation in this field.

EGR1 is a gatekeeper of inflammatory enhancers in human macrophages

Marco Trizzino et al.

Summary: Analyzing the EGR1 transcription factor can reveal the molecular determinants of monocytic and macrophagic commitment, with EGR1 being essential for monopoiesis but showing a different binding pattern in differentiating macrophages. The repressive activity of EGR1 on inflammatory genes is mediated by the NuRD corepressor complex, even in the absence of its binding motif.

Transcriptional analysis of islets of Langerhans from organ donors of different ages

Peter Seiron et al.

Summary: The study showed that genes related to aging are upregulated with age, while several aspects of the cell cycle machinery are downregulated. Additionally, many genes not previously linked to aging were identified, possibly due to different methods of isolating islet cells in earlier studies.

BBS7-SHH Signaling Activity Regulates Primary Cilia for Periodontal Homeostasis

Pi En Chang et al.

Summary: The study found that the expression of BBS7 was downregulated in occlusal hypofunctional PDL tissue. Occlusal hypofunction led to dynamic changes in PDL, affecting the number and alignment of cells, blood vessel diameter, appearance of primary cilia, and structure of oxytalan fibers. Additionally, BBS7 was found to be closely associated with Sonic hedgehog signaling and played a key role in maintaining PDL homeostasis.

Hyperglycemia modulates M1/M2 macrophage polarization via reactive oxygen species overproduction in ligature-induced periodontitis

Bo Zhang et al.

Summary: The study found that high glucose could polarize macrophages toward M1 macrophages via overproducing ROS, which may be responsible for aggravated periodontal damage in periodontitis under diabetic condition. Inhibiting M1 macrophages and restoring M2 macrophages by ROS scavenger is potentially a adjunct treatment strategy for diabetic periodontitis.
