Prolyl 4-hydroxylase subunit alpha 3 facilitates human colon cancer growth and metastasis through the TGF-beta/Smad signaling pathway

Hailang Zhou et al.

Summary: The study showed that P4HA3 promotes human colon cancer growth and metastasis by affecting the TGF-beta/Smad signaling pathway. P4HA3 knockdown significantly decreased migration, proliferation, and invasion abilities of colon cancer cells, while overexpression had the opposite effect. Additionally, P4HA3 can serve as a potential new target for early diagnosis, treatment, and prognosis assessment of colon cancer.

Microenvironmental sensing by fibroblasts controls macrophage population size

Xu Zhou et al.

Summary: This article reports on the mechanisms controlling the population numbers of different cell types in animal tissues. The study shows that fibroblasts are limited in proliferation due to space availability, while macrophages are constrained by growth factor availability. The transcriptional coactivator YAP1 in fibroblasts directly regulates the expression of the macrophage-specific growth factor Csf1. The findings suggest that tissue environment sensing is a general strategy for controlling tissue composition.

Aging and Susceptibility to Pulmonary Disease

Julia Budde et al.

Summary: The lungs are vulnerable to diseases of aging due to continuous exposure to harmful substances and immune activity. Aging results in cellular defects and tissue dysfunction due to the accumulation of unrepaired random molecular damage over time. The breakdown of cellular mechanisms accelerates the aging process and contributes to diseases such as COPD, IPF, and cancers. Aging weakens the immune system and leads to chronic inflammation called inflammaging. The respiratory system undergoes expected changes, including a loss of lung elasticity, decrease in muscle strength, ventilation-perfusion mismatching, and vascular stiffening. Recognizing comorbidities and understanding the differences in pulmonary disease presentation between elderly and younger adults can improve clinical outcomes.

Single-cell transcriptomic atlas of primate cardiopulmonary aging

Shuai Ma et al.

Summary: The research indicates that aging is associated with increased systemic inflammation and compromised virus defense in the cardiopulmonary system. With age, expression of the SARS-CoV-2 receptor ACE2 increases, while the accumulation of the cytokine IL7 induces ACE2 expression. Treatment with vitamin C can block IL7-induced ACE2 expression, suggesting that geroprotective strategies may reduce the severity of COVID-19 in the elderly.

The aging transcriptome and cellular landscape of the human lung in relation to SARS-CoV-2

Ryan D. Chow et al.

Summary: Age is a major risk factor for severe illness from COVID-19, and the heightened severity of COVID-19 in older populations may be related to age-associated changes in the transcriptome and cellular landscape of the aging lung. Specific age-associated cells and genes interact with the SARS-CoV-2 proteome and are dysregulated by SARS-CoV-2 infection in vitro and in patients with severe COVID-19.

Molecular programs of fibrotic change in aging human lung

Seoyeon Lee et al.

Summary: Analysis of lung samples from healthy human donors reveals an increase in cellular senescence and pro-fibrotic pathway activation with age. Telomere shortening, decreased lung epithelial cells, and increased fibroblasts are associated with aging. This study shows a profile of increased vulnerability to lung diseases with age, highlighting the importance of genomic analysis and live lung imaging in understanding the natural history of human lung aging.

IL-6 in inflammation, autoimmunity and cancer

Toshio Hirano

Summary: IL-6 is involved in various physiological processes including immune responses and inflammation. It plays a crucial role in chronic inflammation and cytokine storms, such as seen in COVID-19. The IL-6 amplifier, involving NF-kappa B and STAT3, is a key mechanism in promoting uncontrolled inflammatory responses.
