Omics-based identification of an NRF2-related auranofin resistance signature in cancer: Insights into drug repurposing

Marcelo Falchetti et al.

Summary: This study characterizes the transcriptional and genetic alterations associated with auranofin response in cancer cells. It identifies a signature of 29 genes that are associated with auranofin resistance, with mutations in NRF2 pathway genes strongly linked to overexpression of these genes. The study also predicts the sensitivity of different cancer types to auranofin based on their gene expression patterns.

Interferon-γ induces retinal pigment epithelial cell Ferroptosis by a JAK1-2/STAT1/SLC7A11 signaling pathway in Age-related Macular Degeneration

Ting-Ting Wei et al.

Summary: The study shows that IFN-gamma induces ferroptosis in ARPE-19 cells by activating the JAK1-2/STAT1/SLC7A11 signaling pathway, leading to AMD progression. Inhibiting ferroptosis or reducing IFN-gamma may be a promising therapeutic target for AMD.

The Relationship of Redox With Hallmarks of Cancer: The Importance of Homeostasis and Context

Faliang Xing et al.

Summary: Redox homeostasis plays a crucial role in the development of cancer cells, with reactive oxygen species (ROS) exerting paradoxical effects. In an appropriate concentration, ROS stimulates tumorigenesis and supports cancer cell progression, while excessive concentration leads to cell death. Cancer cells upregulate their antioxidant system to limit ROS to a level that promotes tumor growth. Redox regulation interacts with various aspects of cancer, including tumor initiation, proliferation, metastasis, programmed cell death, autophagy, metabolic reprogramming, the tumor microenvironment, therapies, and therapeutic resistance.

Targeting pan-essential genes in cancer: Challenges and opportunities

Liang Chang et al.

Summary: Despite significant successes in clinical settings, the development of cancer targeted therapy remains challenging, with a disappointingly high failure rate. The misapplication of the targeted therapy paradigm to therapeutics targeting pan-essential genes is a key factor contributing to this issue. Strategies to avoid previous pitfalls and support the ongoing and future development of pan-essential therapeutics are suggested.

Ferroptosis: molecular mechanisms and health implications

Daolin Tang et al.

Summary: Cell death can be executed through different subroutines, such as ferroptosis which is an iron-dependent form of non-apoptotic cell death. Ferroptosis can occur through two major pathways, driven by redox imbalance and abnormal expression of redox-active enzymes. The process is precisely regulated at multiple levels, with the transcription factor NFE2L2 playing a central role in anti-ferroptotic defense.

Comprehensive Review of Methodology to Detect Reactive Oxygen Species (ROS) in Mammalian Species and Establish Its Relationship with Antioxidants and Cancer

Shivkanya Fuloria et al.

Summary: Evidence suggests that reactive oxygen species (ROS) play complex roles in cancer, with both beneficial and harmful effects, depending on concentration. Further research is needed to understand the relationship between ROS, antioxidants, and cancer, and to develop more accurate methods for ROS detection.

Broadening horizons: the role of ferroptosis in cancer

Xin Chen et al.

Summary: Ferroptosis is an iron-dependent form of regulated cell death driven by excessive lipid peroxidation, and can be induced by various agents to suppress tumor growth while also potentially triggering inflammation-associated immunosuppression. The extent of ferroptosis's impact on tumor biology and its interactions with other signaling pathways are still under investigation.

SC75741, A Novel c-Abl Inhibitor, Promotes the Clearance of TDP25 Aggregates via ATG5-Dependent Autophagy Pathway

Dongheng Zhou et al.

Summary: The study demonstrates that SC75741 exerts neuroprotective effects on ALS through multiple mechanisms such as inhibiting NF-κB, enhancing TFEB nuclear translocation, and promoting TDP25 degradation. Additionally, the dual-targeted inhibition of c-Abl and NF-κB by SC75741 may be a potential therapeutic approach for TDP43 proteinopathies and ALS.

Summary: Copper plays a key role in cancer progression and copper-based complexes are being investigated as novel anticancer drugs. The combination of DSF with copper has shown great anticancer potential, but DSF has proven unsuccessful in clinical trials, likely due to poor stability of the currently used oral version of DSF.

Proteogenomics of non-small cell lung cancer reveals molecular subtypes associated with specific therapeutic targets and immune-evasion mechanisms

Janne Lehtio et al.

Summary: Researchers conducted a proteogenomic analysis of non-small cell lung cancer, identifying molecular subtypes with distinct immune-evasion mechanisms and therapeutic targets. They validated their classification method in separate clinical cohorts, discovering high neoantigen burden linked to global hypomethylation and immune evasion mechanisms associated with STK11 mutation-dependent HNF1A activation and FGL1 expression. Furthermore, they developed a data-independent acquisition mass-spectrometry-based NSCLC subtype classification method, validating it in independent cohorts and demonstrating clinical utility.
