From adenoma to CRC stages: the oral-gut microbiome axis as a source of potential microbial and metabolic biomarkers of malignancy

Tissue vs. Fecal-Derived Bacterial Dysbiosis in Precancerous Colorectal Lesions: A Systematic Review

Summary: While alterations in intestinal bacterial microbiota are recognized as important factors in colorectal carcinogenesis, the links between microbiota changes and premalignant colorectal polyps have not been fully examined. This systematic review aims to assess the differences between tissue-derived and stool-based bacterial dysbiosis in patients with colorectal adenoma and carcinoma in situ and propose the correct selection of each matrix for future microbiota studies. The majority of included studies associate gut microbial dysbiosis with premalignant polyps, and there are common characteristics in stool-based and fecal-derived gut microbiota structures in patients with colorectal polyps. Mucosal samples are more relevant for evaluating microbiota's involvement in colorectal carcinogenesis, while non-invasive stool sampling could be beneficial for early CRC detection strategies.

The Application of Metabolomics in Recent Colorectal Cancer Studies: A State-of-the-Art Review

Summary: Colorectal Cancer (CRC) is a leading cause of cancer-related death in the United States and current diagnosis techniques lack sufficient sensitivity. Metabolomics is a promising technique for identifying CRC biomarkers with high sensitivity and specificity, but it is rarely used in clinical settings. This study conducted a systematic review of recent literature on the detection of CRC biomarkers through metabolomics, identifying dysregulated metabolites across different biospecimens. The study also highlighted the interplay between the gut microbiome and perturbed metabolites in CRC. The findings demonstrate the value of metabolomics as a promising diagnostic technique for CRC.

Summary: This study found significant associations between Parvimonas micra and high-grade tumors as well as tumors of CMS1 subtype, and significant associations between Fusobacterium nucleatum and right-sided tumors, microsatellite instability, and CMS1 tumors. Parvimonas micra was associated with activated T lymphocytes and antigen-presenting B lymphocytes, while no associations were found between Fusobacterium nucleatum and immune profiles in this study.

Integrated analysis of the faecal metagenome and serum metabolome reveals the role of gut microbiome-associated metabolites in the detection of colorectal cancer and adenoma

Summary: By integrating untargeted serum metabolomics and metagenome sequencing, this study identified 8 gut microbiome-associated serum metabolites that accurately distinguished CRC and adenoma. The model based on these metabolites showed high diagnostic accuracy for CRC and adenoma, outperforming traditional clinical markers like carcinoembryonic antigen. This suggests that gut microbiome-associated metabolites have potential applications in detecting CRC and adenoma.

Analysis of changes in microbiome compositions related to the prognosis of colorectal cancer patients based on tissue-derived 16S rRNA sequences

Summary: This study analyzed microbiome compositions in tumor and normal tissues of CRC patients, and revealed that certain pathogenic bacteria were more abundant in tumor tissues. Additionally, bacteria associated with bacterial motility proteins and bile acid secretion pathways were enriched in tumor tissues. Interestingly, the abundance of pathogenic bacteria was positively correlated with the expression of host genes relevant to cell cycle and proliferation. Moreover, the presence of pathogenic bacteria was unexpectedly higher in nonrecurrent CRC patients compared to those with recurrence.

Metabolic profiling analysis for clinical urine of colorectal cancer

Summary: This study aimed to demonstrate the metabolic changes and pathways in patients with colorectal cancer through urine metabolic profiling using GC-TOF/MS. The results showed clear discriminations in metabolic profiles between CRC patients and healthy controls, with significant upregulation of glycolysis and amino acid metabolism and downregulation of lipid metabolism in CRC patients. GC-TOF/MS-based metabolomics has the potential to be a novel, non-invasive tool for CRC diagnosis and understanding disease mechanisms.
