Characterization of Selenium Compounds for Anti-ferroptotic Activity in Neuronal Cells and After Cerebral Ischemia-Reperfusion Injury

Qing-Zhang Tuo et al.

Summary: The emergence of ferroptosis as a cell death pathway associated with brain disorders highlights the importance of developing therapeutics targeting the brain and protecting neurons. Selenium compounds have shown to be effective in inhibiting ferroptosis, with organic selenium compounds like methylselenocysteine providing protection against neuronal damage in ischemia-reperfusion injury in vivo. This suggests the potential use of ferroptosis inhibitors and specific selenium compounds in preventing neuronal damage in ischemic stroke and other related brain diseases.

NAD plus supplementation reduces neuroinflammation and cell senescence in a transgenic mouse model of Alzheimer's disease via cGAS-STING

Yujun Hou et al.

Summary: Research reveals that levels of NAD* decrease, and inflammation increases in the brains of Alzheimer's disease patients, while treatment with NR can increase brain NAD* levels, reduce inflammation, and improve cellular senescence. NR treatment is beneficial for neuroinflammation and cellular senescence through a cGAS-STING dependent pathway, inducing mitophagy and improving cognitive function.

Role of endolysosome function in iron metabolism and brain carcinogenesis

Peter W. Halcrow et al.

Summary: Iron is a crucial microelement in the human brain, playing essential roles in regulating cellular mechanisms. However, imbalanced iron homeostasis can lead to accumulation of excess reactive oxygen species in endolysosomes, causing damage to mitochondria and DNA, ultimately promoting carcinogenesis. Understanding the impact of endolysosome iron accumulation on cancer development may aid in the development of effective therapeutic strategies for cancer treatment and prevention.

Inhibition of IP3R/Ca2+Dysregulation Protects Mice From Ventilator-Induced Lung Injury via Endoplasmic Reticulum and Mitochondrial Pathways

Liu Ye et al.

Summary: Disruption of intracellular calcium homeostasis is implicated in inflammatory responses. High tidal volume mechanical ventilation leads to increased IP3R expression, disrupted Ca2+ homeostasis, and inflammatory responses. Targeting IP3R/Ca2+ or maintaining Ca2+ homeostasis may be a potential treatment approach for ventilator-induced lung injury.

Summary: The study demonstrated that activated microglia in the motor cortex induced iron overload after SCI, triggering motor neuronal ferroptosis and impeding motor functional recovery. Iron chelator and inhibitors could reduce iron overload-induced neuronal death, potentially offering novel therapeutic strategies for SCI.
