Glioma targeted therapy: insight into future of molecular approaches

Keyang Yang et al.

Summary: Gliomas are common brain tumors originating from glial cells. Treatment and prognosis differ significantly depending on the type of glioma. Traditional targeted therapy approaches have faced challenges, while new methods such as immunotherapy and the tumor microenvironment are gaining interest. More clinical trials are needed to evaluate the feasibility of personalized targeted therapy.

SMGR: a joint statistical method for integrative analysis of single-cell multi-omics data

Qianqian Song et al.

Summary: In this study, a novel method called SMGR was developed to detect functional regulatory signals and target genes from single-cell multi-omics data. Results showed that SMGR outperformed existing methods in terms of accuracy. Application of SMGR to mixed-phenotype acute leukemia (MPAL) identified MPAL-specific regulatory programs, enhancing our understanding of the regulatory mechanisms and potential targets of this complex tumor.

Regulation of branched-chain amino acid metabolism by hypoxia-inducible factor in glioblastoma

Bo Zhang et al.

Summary: This study reveals that hypoxia induces upregulation of the BCAA transporter LAT1 and the BCAA metabolic enzyme BCAT1 in human glioblastoma cells through the mediation of HIF-1 and HIF-2. HIF-1α plays a critical role in hypoxia-induced BCAT1 expression, and knockout of HIFs reduces glutamate labeling from BCAAs in GBM cells. Inhibition of BCAT1 inhibits GBM cell growth under hypoxia, highlighting a previously unrecognized HIF-dependent metabolic pathway.

Immune suppression in gliomas

Matthew M. Grabowski et al.

Summary: Gliomas evade immune detection and destruction through mechanisms such as dysfunctional T cells and NK cells, expansion of regulatory T cells and myeloid-derived suppressor cells, M2 phenotypic transformation in macrophages/microglia, upregulation of immunosuppressive factors, tumor microenvironment hypoxia, and iatrogenic effects of immunosuppressive treatments. Future research should focus on addressing these immunosuppressive mechanisms to develop treatment options with improved survival outcomes.

Summary: SDT shows promise as a treatment for malignant gliomas, combining low-intensity ultrasound waves with sensitizing compounds to enhance immunogenicity, increase apoptotic rates, and reduce angiogenetic potential in tumor cells. Preclinical studies have confirmed its anti-tumoral properties and first clinical trials are set to begin recruiting patients. Further investigations are needed to explore the clinical applications of SDT for intracranial gliomas.

Reduction of Bladder Cancer Chemosensitivity Induced by the Effect of HOXA-AS3 as a ceRNA for miR-455-5p That Upregulates Notch1

Dajin Chen et al.

Summary: HOXA-AS3 functions as a competing endogenous RNA (ceRNA) of miR-455-5p to regulate Notch1 and play a crucial role in chemoresistance in bladder cancer cells. Inhibition of HOXA-AS3 can enhance sensitivity to cisplatin in bladder cancer cells, providing a potential new therapeutic target for treating bladder cancer.

Sensing and Signaling of Methionine Metabolism

Linda Lauinger et al.

Summary: The availability of the amino acid methionine plays a crucial role in the physiology of individual cells and whole organisms, particularly in cancer cells and healthy lifespan. Cells need mechanisms to sense methionine availability, and respond through transcriptional and signaling states to maintain metabolic balance and genomic integrity during methionine starvation conditions.

Altered Metabolism in Glioblastoma: Myeloid-Derived Suppressor Cell (MDSC) Fitness and Tumor-Infiltrating Lymphocyte (TIL) Dysfunction

Natalia Di Ianni et al.

Summary: Glioblastoma metabolism is flexible and adaptable to adverse conditions, with altered lipid metabolism also playing a role in tumor progression. Immunosuppressive microenvironment of GBM involves MDSCs as allies of tumor cells, and there is increasing evidence of interconnection between GBM and MDSC metabolic pathways. Understanding the metabolic programs of GBM and MDSCs is important for improving treatment modalities, including immunotherapeutic strategies.

SYVN1-MTR4-MAT2A Signaling Axis Regulates Methionine Metabolism in Glioma Cells

Lude Wang et al.

Summary: This study investigated the molecular mechanism of glioma response to methionine restriction, revealing that under methionine restriction, glioma cells showed high expression of MAT2A, and an inhibitor of MAT2A reduced the proliferation of tumor cells. High expression of MAT2A was associated with World Health Organization-grade glioma and increased transfer of its mRNA out of the nucleus.
