IFNγ binding to extracellular matrix prevents fatal systemic toxicity

Summary: The study demonstrates that the extracellular matrix (ECM)-binding domain (EBD) of interferon-gamma (IFN gamma) is crucial for preventing high systemic levels of this cytokine and related immunopathology. Lack of EBD in IFN gamma significantly reduces ECM binding but retains the binding to IFN gamma receptor and bioactivity. Overexpression of EBD-deficient IFN gamma results in increased systemic levels, sickness behavior, weight loss, and toxicity.

Loss of the Sympathetic Signal Produces Sterile Inflammation of the Prostate

Summary: This study used advanced techniques to investigate the anatomical characteristics of autonomic innervations in the mouse and human prostate for the first time. The research found that sympathetic and parasympathetic inputs in the mouse prostate remained unaffected during castration-induced tissue regression. However, the destruction of sympathetic innervations led to sterile inflammation, mimicking the disease condition of chronic non-bacterial prostatitis. Treatment with specific beta 2-adrenergic receptor agonists effectively mitigated the prostate inflammation caused by sympathetic loss.
