Dopamine-2 receptor antibody encephalitis presenting as pure tongue-biting in a tourette syndrome patient: a case report

Mingfeng Lai et al.

Summary: This article reports a case of D2R encephalitis in a patient with Tourette syndrome (TS). The patient presented with tic symptoms, but significant improvement was observed after early immunotherapy. This suggests that clinicians should be aware of D2R encephalitis in TS patients and consider early immunotherapy based on clinical characteristics.

Interleukin-4 Aggravates LPS-Induced Striatal Neurodegeneration In Vivo via Oxidative Stress and Polarization of Microglia/Macrophages

Jaegeun Jang et al.

Summary: This study investigated the effects of IL-4 on striatal neurons in LPS-injected rat striatum. The results showed a significant loss of striatal neurons and increased IL-4 immunoreactivity post LPS injection. The use of IL-4 neutralizing antibody protected striatal neurons against LPS-induced neurotoxicity and inhibited activation of microglia/macrophages and oxidative damage. It also resulted in a shift of microglia/macrophages from M1 to M2 polarization.

Boosted dopamine and blunted serotonin in Tourette syndrome - evidence from in vivo imaging studies

Susanne Nikolaus et al.

Summary: This study found that medication-naive patients with Tourette syndrome showed increased binding of DA transporter in certain regions of the brain, while D-2 receptor binding and DA release were not different from controls. When patients were medication-free, the binding of DAT was still increased in some regions, while other regions showed no difference. SERT was decreased in certain regions and 5-HT2AR binding was increased in certain regions. Acutely medicated patients showed no change in DAT in certain regions, while other regions still showed increased binding.

Profiles of Proinflammatory Cytokines and T Cells in Patients With Tourette Syndrome: A Meta-Analysis

Ying Li et al.

Summary: This study conducted a meta-analysis to investigate immune dysfunction in pediatric TS patients, focusing on proinflammatory cytokines and T-cell changes. The results showed elevated levels of specific proinflammatory cytokines and disproportionate changes in T-cell subpopulations in TS patients. Increased IL-6 levels and a reduced number of T helper cells were identified with small to large effect sizes, while increased TNF-alpha levels had a large effect size. These findings indicate a close association between peripheral immune activation and TS, but further exploration is needed to understand the direct and meaningful interaction between peripheral immune status and microglial activation in the central nervous system in TS patients.

Interleukin 13 on Microglia is Neurotoxic in Lipopolysaccharide-injected Striatum in vivo

Ah Reum Hong et al.

Summary: This study reveals that LPS injection leads to neuronal loss and microglial activation in the striatum, with IL-13 playing a crucial role in this process. Increased IL-13 expression is associated with astrocyte loss and BBB disruption, and treatment with IL-13 neutralizing antibody can protect neurons from LPS-induced neurotoxicity. Interestingly, IL-13NA treatment also promotes the production of neurotrophic factors in the LPS-injected striatum, providing protection for astrocytes.

Modulating Microglia/Macrophage Activation by CDNF Promotes Transplantation of Fetal Ventral Mesencephalic Graft Survival and Function in a Hemiparkinsonian Rat Model

Kuan-Yin Tseng et al.

Summary: Parkinson's disease is a neurodegenerative disorder characterized by the loss of dopaminergic neurons. Cell transplantation is a potential therapy for Parkinson's disease, but it is limited by immune reactions. This study investigates the effect of cerebral dopamine neurotrophic factor (CDNF) on immune responses and the success of cell transplantation in a Parkinson's disease animal model. The results show that CDNF administration enhances the survival of grafted dopaminergic neurons and improves functional recovery. The findings suggest that CDNF may be used to improve transplantation-based regenerative therapies.

Microglia involvement in sex-dependent behaviors and schizophrenia occurrence in offspring with maternal dexamethasone exposure

Chan Rim et al.

Summary: Fetal microglia, being sensitive to changes in the utero environment, may play a role in the sex-biased onset of psychiatric disorders. In this study, prenatal exposure to dexamethasone induced schizophrenia-relevant behaviors in male mice and depressive-like behavior in female mice. The altered microglial function and dopaine system proteins were observed in the affected brain regions. These findings suggest that altered microglial function and increased synaptic pruning may be involved in the occurrence of psychiatric disorders in the prenatal dexamethasone-exposed offspring.

How microglia sense and regulate neuronal activity

Anthony D. Umpierre et al.

Summary: Microglia are innate immune cells in the central nervous system that sense extracellular cues and exhibit dynamic structural responses in brain injuries, inflammation, and pathology. Their dynamic motility is hypothesized to be crucial for sensing local alterations and engaging in pattern-specific responses, as well as regulating neuronal activity. Emerging in vivo evidence suggests that microglia play a role in monitoring and influencing neuronal activity, with calcium signaling also playing a part in their responses to the extracellular environment in health and disease.

Microglial activation elicits a negative affective state through prostaglandin-mediated modulation of striatal neurons

Anna M. Klawonn et al.

Summary: The activation of microglia in neurological diseases, especially in major depression, has been found to be crucial in inducing negative affective states. This is mediated by the microglial production of cytokines and prostaglandins, leading to alterations in the excitability of striatal neurons. Intervention targeting this mechanism could potentially alleviate depressive symptoms in diseases involving microglial activation.
