Summary: This study aimed to determine the mechanisms of VEA-related lung injury and the potential additive lung injury caused by viral infection in the presence of aerosolized VEA. The results showed that extended vaping did not worsen lung injury but caused systemic inflammation. Vaping plus influenza infection increased lung water compared to virus alone. Alveolar macrophages exposed to vaped VEA hydrolyzed the VEA to vitamin E, resulting in oxidative stress in the lungs and systemic circulation. Aerosolized VEA also induced cell death and chemokine release and reduced efferocytotic function in human alveolar macrophages in vitro.

Summary: Electronic nicotine delivery systems (ENDS) have rapidly evolved and pose unique challenges and opportunities for researchers and regulators. This commentary emphasizes the importance of toxicity research and highlights the gaps in knowledge in this area. Disposable flavored ENDS have become popular among youth and may contain higher levels of nicotine compared to JUUL devices. They also produce harmful and potentially harmful constituents and raise environmental concerns. Synthetic nicotine and flavor capsules have allowed manufacturers to bypass regulatory oversight. Coil-less ENDS offer potential advantages in terms of reduced toxicants, but initial studies show similar toxicological profiles to JUULs. Further research is needed to understand and address the impact of these devices on human health, particularly among young users.
