Immune response following traumatic spinal cord injury: Pathophysiology and therapies

Robert C. Sterner et al.

Summary: Traumatic spinal cord injury (SCI) is a devastating condition that leads to loss of function and disability. The pathophysiology involves immediate cell dysfunction and death followed by secondary injury mechanisms, with inflammation playing a key role. Targeting the immune response after SCI could effectively reduce secondary injury and improve outcomes. Recent studies have provided molecular insights into the timing of the immune response and potential therapies such as pharmaceutical interventions, stem cell therapy, and extracellular vesicle therapy.

SU16f inhibits fibrotic scar formation and facilitates axon regeneration and locomotor function recovery after spinal cord injury by blocking the PDGFRβ pathway

Ziyu Li et al.

Summary: The study found that the expression of PDGFD and PDGFB gradually increased after spinal cord injury (SCI). PDGFB was mainly secreted by astrocytes, while PDGFD was mainly secreted by macrophages/microglia and fibroblasts. Activation of the PDGFR beta pathway can directly induce fibrotic scar formation. Specific blockade of this pathway contributes to the treatment of SCI.

Microglial activation elicits a negative affective state through prostaglandin-mediated modulation of striatal neurons

Anna M. Klawonn et al.

Summary: The activation of microglia in neurological diseases, especially in major depression, has been found to be crucial in inducing negative affective states. This is mediated by the microglial production of cytokines and prostaglandins, leading to alterations in the excitability of striatal neurons. Intervention targeting this mechanism could potentially alleviate depressive symptoms in diseases involving microglial activation.

The thrombin receptor modulates astroglia-neuron trophic coupling and neural repair after spinal cord injury

Ha Neui Kim et al.

Summary: The study demonstrates that genetic blockade of PAR1 in female mice results in improvements in sensorimotor co-ordination after thoracic spinal cord lateral compression injury. These improvements are accompanied by enhanced neuron preservation, myelin resiliency and repair, as well as reduced pro-inflammatory markers in astrocytes and microglia/monocytes. The loss of PAR1 function in astrocytes promotes both neuron survival and neurite outgrowth, highlighting its role in regulating glial-neuron interactions and neurotrophic factor signaling for CNS injury response and regenerative repair.

Topoisomerase I inhibition and peripheral nerve injury induce DNA breaks and ATF3-associated axon regeneration in sensory neurons

Summary: This study shows that topoisomerase inhibitors, including camptothecin, can induce ATF3 expression, promote neurite outgrowth in sensory neurons, and enhance axonal regeneration. DNA breaks occur immediately after nerve damage at the ATF3 gene locus in injured sensory neurons and are further increased after camptothecin exposure, potentially contributing to the initiation of transcriptional changes required for peripheral nerve regeneration.
