The androgen receptor-lncRNASAT1-AKT-p15 axis mediates androgen-induced cellular senescence in prostate cancer cells

Kimia Mirzakhani et al.

Summary: The study identified that supraphysiological androgen levels induce cellular senescence in prostate cancer cells through the activation of p15(INK4b) and AKT phosphorylation, with lncRNASAT1 playing a crucial regulatory role. This novel AR-lncRNASAT1-AKT-p15(INK4b) signaling axis mediates supraphysiological androgen-induced cellular senescence.

Therapy-Induced Stromal Senescence Promoting Aggressiveness of Prostate and Ovarian Cancer

Elisa Pardella et al.

Summary: Cancer progression is supported by the interaction between tumor cells and the surrounding stroma. Chemotherapy-induced senescence in both tumor and non-cancerous cells contributes to the adverse effects of therapies. It has been found that chemotherapy drugs promote senescence in stromal fibroblasts, leading to metabolic alterations and secretion of paracrine factors, thereby enhancing the invasive and clonogenic potential of prostate and ovarian cancer cells. Clearance of senescent stromal cells can reverse the malignant phenotype of tumor cells. This has important implications for combining traditional anticancer strategies with innovative senotherapy to enhance anticancer treatments and overcome the adverse effects of chemotherapy.

Yuliang Wang et al.

Summary: This study identified ferroptosis-related hub genes associated with disease-free survival (DFS) in prostate cancer (PCa) patients and developed a novel FRGPI as an independent prognostic factor. Patients with high FRGPI scores had worse DFS, and high FRGPI scores were correlated with poor clinicopathological features, higher mutation counts, increased CNVs, higher HRD and immune scores, higher mRNAsi, as well as enhanced sensitivity to immunotherapy.

The olfactory receptor OR51E2 activates ERK1/2 through the Golgi-localized Gβγ-PI3Kγ-ARF1 pathway in prostate cancer cells

Xin Xu et al.

Summary: The olfactory receptor OR51E2 is ectopically expressed in prostate tissues and regulates prostate cancer progression. A study demonstrated that beta-ionone, an OR51E2 agonist, activates ERK1/2 pathway in prostate cancer cells. The activation of ERK1/2 by beta-ionone is regulated by Golgi-translocating G gamma 9 subunit, PI3K gamma, and ARF1.

Identification of senescence-related subtypes, establishment of a prognosis model, and characterization of a tumor microenvironment infiltration in breast cancer

Yanling Zhou et al.

Summary: This study identified different molecular subtypes of breast cancer based on the expression profiles of cellular senescence-related genes (SRGs) and established a senescence-related scoring signature that can predict the prognosis of breast cancer patients. The accuracy of this signature was validated and it was found to be associated with pathological response rate. The study also revealed the potential roles of these senescence patterns in the tumor-immune-stromal microenvironment.

P16INK4A-More Than a Senescence Marker

Hasan Safwan-Zaiter et al.

Summary: Aging is a biological process characterized by the gradual decline in cellular, tissue, organ, or organismal function due to the accumulation of environmental factors and stresses. Several factors contribute to aging, including oxidative stress, inflammatory cytokines production, DNA damage, and the presence of senescent cells. P16(INK4A), a tumor suppressor gene and cell cycle regulator, plays a crucial role in aging and cellular senescence. Extensive research has shown that p16 is not only a specific biomarker for detecting senescent cells and aging, but also has other functions outside of senescence and aging.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
