Mas-related G protein-coupled receptor MRGPRX2 in human basophils: Expression and functional studies

Alessandro Toscano et al.

Summary: This study explored the expression and functionality of MRGPRX2 in and on basophils. The results showed that basophils barely express MRGPRX2 in resting conditions, but it can be quickly upregulated after stimulation and make cells responsive to MRGPRX2 occupation. These findings provide a model for studying MRGPRX2 agonism or antagonism and the mechanisms associated with IDHRs.

Inhibition of Orai Channel Function Regulates Mas-Related G Protein-Coupled Receptor-Mediated Responses in Mast Cells

Shaswati Chaki et al.

Summary: The calcium release-activated calcium channels (CRAC/Orai) contribute to the activation of mast cells (MCs) mediated by MRGPRX2/B2, and their inhibition may provide a novel approach for modulating SP-induced MC/MRGPRX2-mediated disorders.

Dendritic cell immunoreceptor drives atopic dermatitis by modulating oxidized CaMKII-involved mast cell activation

Xiaoyan Luo et al.

Summary: This study reveals the role of allergens in triggering skin inflammation in patients with atopic dermatitis through the activation of mast cells. The researchers also identified the DCIR/ROS/CaMKII axis as a key regulatory pathway in allergen-induced mast cell activation and inflammatory response. The findings provide insights into the underlying mechanism of atopic dermatitis and suggest potential targets for therapeutic intervention.

Cytokine Stimulation by MRGPRX2 Occurs with Lower Potency than by FcεRI Aggregation but with Similar Dependence on the Extracellular Signal-Regulated Kinase 1/2 Module in Human Skin Mast Cells

Zhao Wang et al.

Summary: In skin mast cells, MRGPRX2 activation can induce cytokines, although less efficiently than Fc epsilon RI activation. The MAPK/ERK pathway plays a key role in cytokine production, with inhibitors of this pathway suppressing cytokine secretion. Overall, MRGPRX2-associated inflammation may be less tissue destructive compared to responses to allergic challenges mediated by Fc epsilon RI.

MRGPRX2-Mediated Degranulation of Human Skin Mast Cells Requires the Operation of Gαi, Gαq, Ca++ Channels, ERK1/2 and PI3K-Interconnection between Early and Late Signaling

Zhao Wang et al.

Summary: The recent discovery of MRGPRX2 explains the mast cell-dependent symptoms independently of Fc epsilon RI-activation. MRGPRX2 signals by activating G proteins, mainly depending on Gαi. Ca++ channels and PI3K are crucial in the process as well.

IL-33 Induces an Antiviral Signature in Mast Cells but Enhances Their Permissiveness for Human Rhinovirus Infection

Charlene Akoto et al.

Summary: In this study, the researchers investigated the response of mast cells (MCs) to IL-33 stimulation and found that it increased the susceptibility of MCs to HRV infection through upregulation of ICAM1. This highlights a potential gene-environment interaction that could contribute to virus-induced asthma exacerbations.

The high affinity IgE receptor: a signaling update

Ulrich Blank et al.

Summary: This article summarizes the receptor proximal and distant signaling events of the mast cell high affinity IgE receptor (FceRI) and its role in launching immediate type I hypersensitivity and an inflammatory cytokine-chemokine cascade. It emphasizes the impact of different physiologic antigen concentrations, affinity, and valency for the IgE ligand, as well as recent research progress in mast cell degranulation.

Thymic Stromal Lymphopoietin Promotes MRGPRX2-Triggered Degranulation of Skin Mast Cells in a STAT5-Dependent Manner with Further Support from JNK

Magda Babina et al.

Summary: TSLP is a novel factor that primes human skin MCs to support granule discharge, selectively cooperating with MRGPRX2 without impacting other pathways. TSLP prolongs granule opening by activating STAT5 and JNK in skin MCs, leading to reinforced degranulation.

Imperatorin ameliorates mast cell-mediated allergic airway inflammation by inhibiting MRGPRX2 and CamKII/ERK signaling pathway

Nan Wang et al.

Summary: The study revealed that IMP can reduce SP-induced calcium flux and suppress MC degranulation. IMP can alleviate SP-induced mouse PCA and ASA, and mitigate lung inflammation in an OVA-induced asthma model by inhibitiing MC activation in lung tissue. Additionally, IMP binds well to MRGPRX2 protein, suggesting it is a novel inhibitor for treating allergic asthma.

Cytokines Stimulated by IL-33 in Human Skin Mast Cells: Involvement of NF-κB and p38 at Distinct Levels and Potent Co-Operation with FcεRI and MRGPRX2

Kristin Franke et al.

Summary: This study found that IL-33 activates various cytokines in cutaneous MCs through different mechanisms, with NF-kappa B playing a significant role in the process. The research highlights the diversity of cytokine responses to IL-33 in different MC subsets and suggests that IL-33 may synergize with other activation networks to amplify skin inflammation, particularly in the presence of allergens or MRGPRX2 agonists.

Multifaceted MRGPRX2: New insight into the role of mast cells in health and disease

Saptarshi Roy et al.

Summary: MRGPRX2, activated by various cationic ligands, plays a role in host defense but overactivation may lead to pseudoallergy and various skin diseases. Gain and loss-of-function missense polymorphisms have been identified in MRGPRX2, and the functionality of certain ligands has been defined. The development of small-molecule HDP mimetics that activate MRGPRX2 presents a new therapeutic approach, while MRGPRX2-specific antibodies and inhibitors could be utilized for modulation of allergic and inflammatory diseases.
