Immune correlates analysis of the mRNA-1273 COVID-19 vaccine efficacy clinical trial

Peter B. Gilbert et al.

Summary: In the COVE phase 3 clinical trial, neutralizing and binding antibodies were measured in vaccine recipients to determine their correlation with COVID-19 risk and vaccine efficacy. The results showed that these immune markers were inversely associated with COVID-19 risk and directly associated with vaccine efficacy. Postvaccination neutralization titers of 10, 100, and 1000 correlated with estimated vaccine efficacies of 78%, 91%, and 96% respectively.

SARS-CoV-2 BNT162b2 vaccine-induced humoral response and reactogenicity in individuals with prior COVID-19 disease

Steven G. Kelsen et al.

Summary: Individuals with prior COVID-19 disease show a different immune and reactogenicity response to the BNT162b2 vaccine compared to those without prior COVID-19 infection. The COVID-19 group has a stronger and faster immune response after the first dose but a weaker response after the second dose of the vaccine. They also experience more adverse reactions.

Will Host Genetics Affect the Response to SARS-CoV-2 Vaccines? Historical Precedents

Maria K. Smatti et al.

Summary: The emerging field of immunogenomics, which combines immunology and genetics, has shed light on how the immune system responds to infection and vaccination. While extensive research is being conducted on the immunogenetic basis of the clinical variability in response to SARS-CoV-2 infection, the genetic factors influencing response to SARS-CoV-2 vaccines remain largely unknown. Understanding the role of host genetics in vaccine-induced immunity could lead to personalized vaccine strategies to optimize vaccine response.

Variant-specific symptoms of COVID-19 in a study of 1,542,510 adults in England

Matthew Whitaker et al.

Summary: This study assesses changing symptom profiles associated with different SARS-CoV-2 variants in England from May 2020 to March 2022. The authors find that Omicron infection is more often associated with cold and influenza-like symptoms, and less with loss of taste and smell. Contrary to the perception that recent variants have become milder, Omicron BA.2 was associated with reporting more symptoms and greater disruption to daily activities than BA.1.

Correlates of protection against symptomatic and asymptomatic SARS-CoV-2 infection

Shuo Feng et al.

Summary: Defined levels of SARS-CoV-2-specific binding and neutralizing antibodies elicited by the COVID-19 vaccine were identified as correlates of protection against symptomatic infection. Higher levels of immune markers were correlated with a reduced risk of symptomatic infection. The data can be used to extrapolate efficacy estimates to new populations.

Waning Immune Humoral Response to BNT162b2 Covid-19 Vaccine over 6 Months

Einav G. Levin et al.

Summary: A study in Israel revealed that waning immunity after receiving two doses of the BNT162b2 vaccine led to an increase in the incidence of SARS-CoV-2 infection. Levels of spike-binding IgG and neutralizing antibodies decreased more significantly in men, individuals aged 65 or older, and immunosuppressed individuals in a longitudinal study involving nearly 4000 healthcare workers.

Summary: This statewide study in Virginia found that 2.4% of adults had been exposed to SARS-CoV-2, significantly higher among Hispanic individuals, those living in multifamily units, and those in contact with confirmed COVID-19 cases. Most infections were asymptomatic, indicating a largely immunologically naive population in Virginia as of August 2020.

Influence of past infection with SARS-CoV-2 on the response to the BNT162b2 mRNA vaccine in health care workers: Kinetics and durability of the humoral immune response

Jesus Ontanon et al.

Summary: The study shows that at least ten months post-COVID-19 infection, the immune system is still capable of producing a rapid and powerful secondary antibody response following one single vaccine dose. There was a significant increase in antibody levels after the second dose in naive HCWs, while experienced HCWs showed no further improvement in antibody response to the second dose.

Summary: The study found that SARS-CoV-2 seronegative healthcare workers had higher CD4(+) T-cell responses against common cold coronaviruses, possibly due to higher occupational exposure. Additionally, exposure to SARS-CoV-2 may decrease T-cell reactivity against common cold coronaviruses, as observed in independent cohorts from Miami and San Diego.

Dynamics of Neutralizing Antibody Titers in the Months After Severe Acute Respiratory Syndrome Coronavirus 2 Infection

Katharine H. D. Crawford et al.

Summary: Antibody levels, including neutralizing antibodies, decline after SARS-CoV-2 infection, with a 4-fold average decrease in titers from 1 to 4 months after symptom onset. This decline is accompanied by a decrease in total antibodies capable of binding the viral spike protein. Further studies are needed to determine the long-term durability of immunity to SARS-CoV-2, including examination of long-lived B cells and antibody titers over extended periods.
