Evolution of innate defense in human skin

Enno Christophers et al.

Summary: Over millions of years, human skin has undergone fundamental changes in structure and function compared to other barrier systems. The formation of an impermeable stratum corneum and the evolution of a highly developed immune system are key aspects of these changes. After the agricultural revolution, human skin became a battlefield for pathogens, skin microbiome members, and arthropod visits. The keratinocyte-myeloid synergy (KMS) principle plays a vital role in regulating innate immunity and contributes to the development of pustular dermatoses.

Summary: This review discusses the pathogenesis of pediatric IgAVN, highlighting the four-hit hypothesis related to IgAN and the evidence supporting it. It also explores the genetic, clinical outcomes, and histopathological features in pediatric IgAVN, emphasizing the roles of Gd-IgA1-IgG immune complexes and complement activation. Future studies are needed to further investigate the immune responses in the progression from IgA vasculitis to IgAVN.

Complexities of the glomerular basement membrane

Richard W. Naylor et al.

Summary: The glomerular basement membrane (GBM) is crucial for kidney filtration, with studies revealing insights into its composition, assembly, developmental transitions, and filtration role. Defects in GBM components are associated with hereditary and acquired diseases, with current treatments focused on reducing intraglomerular pressure and addressing underlying causes. Therapies targeting GBM maintenance and repair could lead to new treatments for GBM-associated diseases.

IgA Vasculitis and IgA Nephropathy: Same Disease?

Evangeline Pillebout

Summary: Many authors have suggested that IgA Vasculitis and IgA Nephropathy may be two different clinical manifestations of the same disease, but larger clinical studies are needed to explore further. Limited studies have been conducted on these two diseases, and therapeutic trials often exclude patients with both conditions.
