Gnomoniopsis castaneae: An emerging plant pathogen and global threat to chestnut systems

Rapid and Accurate Detection of Gnomoniopsis smithogilvyi the Causal Agent of Chestnut Rot, through an Internally Controlled Multiplex PCR Assay

Summary: In this study, a specific and sensitive multiplex PCR method was developed for the detection of the chestnut nut rot pathogen Gnomoniopsis smithogilvyi. The assay reliability was enhanced by optimizing primer annealing temperature and concentration. The detection limit of the assay was low, and there was no cross-reactivity with closely and distantly related fungal species. Significant differences in morphologic and virulence traits were found among different isolates of G. smithogilvyi.

Metabolites derived from fungi and bacteria suppress in vitro growth of Gnomoniopsis smithogilvyi, a major threat to the global chestnut industry

Summary: This study evaluated the antifungal potential of biological control agents (BCAs) and their secreted metabolites against G. smithogilvyi. The results showed that BCAs significantly suppressed the pathogen's growth through the production of volatile and non-volatile compounds. Metabolomics analysis also revealed the presence of antifungal compounds in the BCAs. These findings suggest that BCAs could be an ecological alternative for managing chestnut rot, but further field studies are needed to assess their effectiveness.

Development of a TaqMan qPCR assay for the detection and quantification of Gnomoniopsis castaneae in chestnut tissues

Summary: A novel real-time PCR assay based on TaqMan probe was developed for sensitive detection and quantification of G. castaneae, the causal agent of brown rot of chestnut kernels. The assay had an amplification efficiency of 105.3% and a limit of detection of approximately 40 fg. It could rapidly detect the pathogen in naturally infected samples, highlighting its potential in detecting G. castaneae in different host tissues.

Machine learning algorithms were used to model propagule deposition patterns and results showed fluctuations driven by temperatures and wind. Climate change may boost the spread of G. castaneae due to increased temperatures and strong winds.
