Cancer statistics, 2022

Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Immunogenic cell stress and death

Guido Kroemer et al.

Summary: Immunogenic cell death (ICD) plays an important role in therapy and disease, as dying mammalian cells release signals that interact with the host to determine the immunological correlates of cellular stress and death. ICD is crucial in immunosurveillance and is related to strategies evolved by pathogens and cancer cells. Additionally, normal cell death can also initiate antigen-specific immune responses.

Restoration of the Immunogenicity of Tumor Cells for Enhanced Cancer Therapy via Nanoparticle-Mediated Copper Chaperone Inhibition

Feixiang Ding et al.

Summary: Recent progress in studying copper-dependent targets and pathways in tumor treatment has provided new insights into leveraging copper-dependent disease vulnerabilities and manipulation of intratumor copper transportation. Researchers developed ROS-sensitive nanoparticles loaded with copper chaperone inhibitor DC_AC50 and cisplatin(IV) prodrug, which enhanced the chemotherapeutic effect of cisplatin, induced significant ROS generation, and facilitated immunogenic cell death (ICD) for sustained immune response.

Summary: The interaction of tumour-associated macrophages (TAMs) with cancer and stromal cells in the tumour microenvironment enables and sustains most of the hallmarks of cancer. The authors of this Review examine the diversity of TAMs in various cancer indications, which is being revisited with the advent of single-cell technologies, and discuss the functional roles of different TAM states, the prognostic and predictive value of TAM-related signatures as well as approaches involving TAMs that are currently being or will soon be tested in clinical trials.

A nuclease-mimetic platinum nanozyme induces concurrent DNA platination and oxidative cleavage to overcome cancer drug resistance

Fangyuan Li et al.

Summary: In this study, a nuclease-mimetic platinum nanozyme was developed to target cancer cell nuclei and induce concurrent DNA platination and oxidative cleavage, overcoming platinum drug resistance. Unlike conventional platinum nanoparticles, the platinum nanozyme specifically cleaves the formed platinum-DNA adducts by generating reactive oxygen species, disrupting the DNA bending required for nucleotide-excision repair. This approach allows for excessive accumulation of platinum-DNA adducts in cisplatin-resistant cancer cells, enabling highly efficient cancer therapy.

Targeting Bone Tumor and Subcellular Endoplasmic Reticulum via Near Infrared II Fluorescent Polymer for Photodynamic-Immunotherapy to Break the Step-Reduction Delivery Dilemma

Xianghong Zhang et al.

Summary: This study presents a cascade targeting NIR II fluorescent nanoparticles that can effectively deliver photosensitizers to bone tumors and subcellular organelles. The delivered photosensitizers generate reactive oxygen species in the organelles under near infrared light irradiation, leading to immunogenic cell death and inhibition of tumor growth. This research provides insights into a safe and efficient nano-drug delivery system with potential clinical applications.

NIR-light triggered dual-cascade targeting core-shell nanoparticles enhanced photodynamic therapy and immunotherapy

Dengshuai Wei et al.

Summary: By utilizing DCT core-shell nanoparticles triggered by near infrared (NIR) light, optimized photodynamic therapy (PDT) and immunotherapy were achieved. These nanoparticles efficiently accumulated at tumor sites and induced robust ROS generation in mitochondria upon continuous light irradiation, leading to cancer cell death via PDT with mitochondrial targeting. These findings highlight a novel strategy for nanoparticles with DCT capability to enhance the effectiveness of combined photodynamic and immunotherapy treatments.

PD-L1 as a biomarker of response to immune-checkpoint inhibitors

Deborah Blythe Doroshow et al.

Summary: PD-L1 expression is currently the best predictor of responsiveness to immune-checkpoint inhibitors, despite challenges in clinical use such as variability in different assays and cut-off points, as well as the lack of prospective comparisons with clinical outcomes. Nevertheless, several immunohistochemical assays have been approved for clinical use, with implications for pathology services and potential impacts on clinical outcomes. Comparison of FDA-approved PD-L1 assays, understanding of the varying implications of PD-L1 expression across different tumour types, and exploration of novel approaches to optimize its clinical utility are important topics for further research and practice.
