Nanostructured 3D-Printed Hybrid Scaffold Accelerates Bone Regeneration by Photointegrating Nanohydroxyapatite

Dopamine-Integrated Nanointerface between Fibrillar Matrix and Hydrophilic Nanohydroxyapatite Regulates Immune Microenvironment to Boost Endogenous Bone Regeneration

Summary: Driving endogenous bone regeneration without the use of cells or factors is the most ideal strategy for repair. A hybrid interleaved scaffold (HDSH) with nanosized interfacial integration is created through organic/inorganic bonding at the nanoscale. Through analysis at the molecular level, it is revealed that this scaffold recreates a suitable immune microenvironment and promotes cell adhesion, cytokine adhesion, and vascularization, leading to enhanced stem cell recruitment and robust vasculogenesis and osteogenesis. In a rabbit cranial defect model, significant bone regeneration is observed after 12 weeks, demonstrating the potential of this strategy for highly bioactive bone-regenerative implants.

The scaffold also exhibited excellent proliferation and differentiation ability of bone marrow mesenchymal stem cells in vitro and bone repair capability in vivo, offering a new idea for bone defect repair with a simple but practical strategy.

3D Bioprinting of Multifunctional Dynamic Nanocomposite Bioinks Incorporating Cu-Doped Mesoporous Bioactive Glass Nanoparticles for Bone Tissue Engineering

Summary: Bioprinting has made significant progress in recent years in the fabrication of complex bionic tissues. However, developing cell-laden bioinks with superior physiochemical properties and bio-functionality remains challenging. In this study, a multifunctional nanocomposite bioink was developed using amine-functionalized copper-doped mesoporous bioactive glass nanoparticles and a hydrogel formulation based on dynamic covalent chemistry. This bioink showed favorable rheological properties, improved shape fidelity, and structural stability for extrusion-based bioprinting. The introduction of aminated particles and the reversible dynamic microenvironment enabled rapid spreading and high survival of embedded cells. The bioprinted scaffolds promoted osteogenic differentiation and angiogenesis of cells without additional growth factors in vitro. These nanocomposite bioinks have great potential for bioprinting complex 3D environments in bone tissue engineering.

Bioinspired Hydrogel Anchoring 3DP GeIMA/HAp Scaffolds Accelerates Bone Reconstruction

Summary: This study developed a biologically inspired bony implant by preparing a 3DP methacrylic anhydride-modified gelatin/hydroxyapatite scaffold with high solid content and anchoring it with a functionalized polyphenol hydrogel. The implant showed improved water retention, delayed degradation, and mechanical stability compared to a simple 3DP scaffold. It also promoted migration, proliferation, and osteogenic differentiation of bone marrow stem cells in vitro, as well as direction migration, early osteogenesis, and angiogenesis in vivo.

Tannin-reinforced iron substituted hydroxyapatite nanorods functionalized collagen-based composite nanofibrous coating as a cell-instructive bone-implant interface scaffold

DLP fabricating of precision GelMA/HAp porous composite scaffold for bone tissue engineering application

Summary: This study aimed to prepare high-precision GelMA/HAp composite scaffolds using digital light processing (DLP) printing for bone repair. GelMA hydrogels with adjustable properties were synthesized, and suitable dispersants were selected to reduce the agglomeration of HAp. The printed composite scaffolds with excellent mechanical strength showed promotion of osteoblast adhesion, proliferation, and differentiation in vitro. In vivo experiments on rabbit skull defects demonstrated the ability of the composite scaffolds to accelerate new bone formation. The GelMA/HAp porous composite scaffolds fabricated by composite inks with good printability, mechanical properties, and osteogenic activity have great potential in bone repair applications.
