Summary: The urgent medical need for novel drugs against Gram-negative bacteria led to the synthesis of antibiotics and artificial siderophores conjugates, with predicted structures and iron binding properties in silico. MECAM demonstrated effective artificial siderophore activity in Escherichia coli, and after linker optimization, certain conjugates showed potent growth inhibition of multidrug-resistant pathogens. The uptake pathway of MECAMs was deciphered, with resistance against one conjugate mediated by a mutation in the gene encoding ExbB involved in siderophore transport.
