Sachin Kumar et al.

Summary: This study discovered a compound that can efficiently bind to the DENV-2 C protein through virtual ligand screening. Molecular docking and dynamics simulations revealed strong interactions between the compound and DENV-2 C protein. The findings suggest that this compound could be a potential therapeutic molecule for treating dengue complications.

Dynamic immune ecosystem of dengue infection revealed by single-cell sequencing

Gang Xu et al.

Summary: Dengue is a common arboviral disease with severe complications. This study characterized the immune infiltrates and dynamic immune ecosystems in dengue infection using single-cell RNA sequencing data. The results showed that genes with higher expression in specific cell types played important roles in response to the virus. Transcription factors were identified as potential regulators of infection-related genes. The study also revealed complex cell-cell communications and identified IFN-II and CXCL signaling pathways involved in dengue infection.

Comprehensive characterization of human-virus protein-protein interactions reveals disease comorbidities and potential antiviral drugs

Si Li et al.

Summary: This study investigates the regulatory pattern of human-virus interactions by analyzing experimentally validated protein-protein interactions (PPIs) between humans and viruses. The research reveals that viral interacting proteins are enriched in cell cycle and immune-related pathways, and tend to locate in central regions of the human PPI network. The study also uncovers associations between complex diseases and viral infections through network proximity analysis and identifies potential antiviral drugs. Additionally, the researchers have developed a Human-Virus Protein-Protein Interaction database that provides experimentally validated human-virus PPIs and functional analysis tools.

Screening and reckoning of potential therapeutic agents against DprE1 protein of Mycobacterium tuberculosis

Rahul Singh et al.

Summary: This study analyzed the interaction ability of aminoarylbenzosuberene (AAB) molecules with DprE1 and identified a promising molecule (AAB4) as a potential lead against tuberculosis. AAB4 showed better inhibition potency and stability compared to co-crystallized inhibitors and also inhibited a drug-resistant mutant. These findings suggest the potential of AAB4 as a therapeutic option for tuberculosis.

Summary: This review article discusses the role of Dengue virus non-structural protein 5 (NS5) and its interactions with other proteins in the human host, particularly focusing on its interference with anti-viral interferon signaling. By annotating and consolidating known and potential NS5 interactors using experimental and computational approaches, the study provides insights for further research efforts to understand how NS5 manipulates the human-virus interface and immune responses.

Innate Lymphoid Cells Activation and Transcriptomic Changes in Response to Human Dengue Infection

Tiraput Poonpanichakul et al.

Summary: Helper innate lymphoid cells (hILCs) are activated during the febrile phase of Dengue virus (DENV) infection, displaying unique transcriptomic changes and cytokine production that correlate with disease severity. Targeting hILCs during the early innate response to DENV may help shape subsequent immune responses and potentially lessen disease severity.
