A novel phage display based platform for exosome diversity characterization

Domenico Maisano et al.

Summary: This study presents an innovative approach to identify, isolate, and characterize disease-related exosomes based on their antigenic reactivities. The strategy can be immediately applied to any disease involving exosomes. The identification of specific markers and their association with exosome subtypes, along with the engineering of target-guided exosome-like particles, could be crucial for the effective clinical adoption of exosomes.

Chemotherapy: a double-edged sword in cancer treatment

Nafiseh Behranvand et al.

Summary: Chemotherapy is a well-known and effective treatment for cancers, but its efficiency in eradicating all cancer cells falls short of expectations. Research suggests that alterations in the tumor microenvironment's physiological conditions may contribute to this. Using mild anti-inflammatory drugs during chemotherapy could potentially lead to more successful clinical outcomes.

Discovery of BT8009: A Nectin-4 Targeting Bicycle Toxin Conjugate for the Treatment of Cancer

Gemma E. Mudd et al.

Summary: Bicycle toxin conjugates (BTCs) are a promising new class of molecules that can deliver toxins to tumor cells effectively. BT8009 is a Nectin-4 targeting BTC that has been optimized through chemical optimization. The optimized bicyclic peptide shows highly selective binding to Nectin-4, good plasma stability, and improved physicochemical properties. The optimized bicycle is conjugated to the cytotoxin Monomethyl auristatin E to form the targeted drug conjugate BT8009, which exhibits potent anticancer activity in vivo rodent models.

Identification of highly selective covalent inhibitors by phage display

Shiyu Chen et al.

Summary: A phage display method was developed to screen for covalent inhibitors that bind to protein targets by introducing a reactive linker to form cyclic peptides and an electrophilic 'warhead' to react with target nucleophiles. This approach identified cyclic peptides that irreversibly inhibited cysteine protease and serine hydrolase with high specificity and potency. It enables rapid, unbiased screening to identify new classes of highly selective covalent inhibitors for diverse molecular targets.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
