Neighboring Carboxylic Acid Boosts Peroxidase-Like Property of Metal-Phenolic Nano-Networks in Eradicating Streptococcus mutans Biofilms

Yaran Wang et al.

Summary: This study reports the development of carboxylic acid-containing nano-networks with enzymatic activity for combating bacterial biofilms. The optimal nano-networks demonstrated high catalytic efficiency in damaging cell membranes and extracellular DNA of Streptococcus mutans biofilms. The neighboring carboxyl group aids the adsorption, free radical generation, and catalyst reactivation, resulting in superb catalytic efficiency. The nano-networks show great potential in inhibiting tooth decay and treating biofilm-associated diseases without affecting the commensal oral flora.

The importance of understanding the infectious microenvironment

Thomas Bjarnsholt et al.

Summary: Standard doses of antibiotics are not effective in treating chronic soft tissue and bone infections. Taking the infectious microenvironment into account could improve the treatment as the microenvironment can cause bacteria to become resistant to antibiotics. Susceptibility testing should be conducted in conditions that resemble the actual infectious microenvironments to guide antibiotic recommendations.

Current In Vitro Biofilm-Infected Chronic Wound Models for Developing New Treatment Possibilities

Ida C. Thaarup et al.

Summary: The prevalence of chronic wounds is increasing globally, with bacterial biofilms identified as a key factor hindering healing. Lack of direct causation and evidence regarding the role of biofilms complicates research on new treatment options, necessitating the use of various in vitro wound models for testing.

Summary: The study found that biosurfactants have effective inhibitory activity against dual-species biofilms, reducing biomass, cell metabolic activity, microstructural architecture, and cell viability. Coating implant surfaces with biosurfactants may be a promising strategy for preventing colonization of Candida and Staphylococcus on medical devices.

Human organoid biofilm model for assessing antibiofilm activity of novel agents

Bing (Catherine) Wu et al.

Summary: A human epidermis organoid model was developed for studying host-microbe interplay and screening for novel antibiofilm agents. Treatment with antibiofilm peptide DJK-5 significantly reduced bacterial burden of MRSA and PAO1 biofilms. Application of DJK-5 on thermally wounded skin infected with MRSA biofilms led to decreased bacterial load, cytotoxicity, and pro-inflammatory cytokine levels.

A novel chronic wound biofilm model sustaining coexistence of Pseudomonas aeruginosa and Staphylococcus aureus suitable for testing of antibiofilm effect of antimicrobial solutions and wound dressings

Xiaofeng Chen et al.

Summary: Chronic wound biofilm infections play a crucial role in hindering wound healing, making it essential to study and develop effective interventions against biofilm. A novel in vitro biofilm model that mimics infected chronic wounds has been introduced, sustaining the coexistence of pathogens for at least 96 hours. Testing with antimicrobial solutions and dressings containing silver or honey showed moderate-to-low antibiofilm effects, aligning better with observed clinical performance compared to standard tests.

Counter-Acting Candida albicans-Staphylococcus aureus Mixed Biofilm on Titanium Implants Using Microbial Biosurfactants

Erica Tambone et al.

Summary: This study aimed to grow a fungal-bacterial mixed biofilm on medical-grade titanium and assess the inhibitory effects of R89BS coating on biofilm formation. Results showed that the R89BS coating significantly inhibited biofilm formation, with over 90% inhibition observed at 24 hours. The coating exhibited similar effects on biofilm biomass, cell metabolic activity, and cell viability, while showing no cytotoxicity on human primary osteoblasts. R89BS coating is a promising strategy to prevent microbial colonization on titanium surfaces, especially for dental implants.
