Vitamin D Supplementation and Fractures in Adults: A Systematic Umbrella Review of Meta-Analyses of Controlled Trials

Marlene Chakhtoura et al.

Summary: This umbrella review investigated the quality and discrepancies of systematic reviews/meta-analyses on vitamin D supplementation for fracture risk reduction in adults. The results show that vitamin D combined with calcium can reduce the risk of hip and any fractures, potentially driven by findings from institutionalized individuals. However, there are discrepancies in results due to different trial inclusion, search periods, eligibility criteria, and methodologies used. Individual participant data meta-analyses and subgroup analysis are recommended to further explore determinants for a beneficial response to supplementation.

Relationship between serum vitamin D and hip fracture in the elderly: a systematic review and meta-analysis

Summary: This study investigated the relationship between serum 25-hydroxyvitamin D (25OHD) levels and the occurrence of hip fractures in the elderly. The results suggest that low serum vitamin D levels are associated with an increased odds of hip fracture.

Summary: The study found that supplementing with vitamin D3 does not significantly reduce the risk of fractures in generally healthy middle-aged and older adults, regardless of total, nonvertebral, or hip fractures.

A comprehensive genetic and epidemiological association analysis of vitamin D with common diseases/traits in the UK Biobank

Summary: Vitamin D has been extensively studied for its association with various diseases and traits in humans. Epidemiological and Mendelian randomization analyses have suggested potential causal relationships between vitamin D and health outcomes, including increased height and reduced risk of certain diseases like ovarian cancer and multiple sclerosis.

Predicting deseasonalised serum 25 hydroxy vitamin D concentrations in the D-Health Trial: An analysis using boosted regression trees

Mary Waterhouse et al.

Summary: The D-Health Trial investigates the impact of high-dose vitamin D supplementation on mortality rate and cancer prevention. Predictive models were developed to estimate baseline serum 25(OH)D levels, with UV radiation and vitamin D intake identified as key predictors of low serum 25(OH)D concentrations.

Vitamin D supplementation and risk of falling: outcomes from the randomized, placebo-controlled D-Health Trial

Mary Waterhouse et al.

Summary: This study found that monthly high-dose vitamin D supplementation did not reduce the risk of falls, and may potentially increase the risk of falling in individuals with normal BMI. Further investigation is needed to better understand the impact of vitamin D supplementation on falls risk in different populations.

Summary: The global incidence, prevalence, and years lived with disability (YLDs) of fractures showed a slight decrease from 1990 to 2019, but the absolute counts increased substantially. Older individuals have a particularly high risk of fractures, highlighting the importance of injury prevention efforts and osteoporosis screening and treatment for reducing the overall burden.
