Aberrant promoter hypermethylation of miR-335 and miR-145 is involved in breast cancer PD-L1 overexpression

Sara Hajibabaei et al.

Summary: PD-L1 is a crucial immune checkpoint molecule in breast cancer, which suppresses the immune system against tumor cells and is regulated by various microRNAs. Among them, microRNA-335-3p and microRNA-145-5p are tumor suppressors regulated by DNA methylation. The downregulation of miR-335 and -145 in breast cancer tissues and cell lines was negatively correlated with PD-L1 overexpression. In addition, the up-regulation of PD-L1 expression was associated with hypermethylation in the CpG-rich areas upstream of miR-335 and-145. Higher levels of miR-355 and -145 induced apoptosis, arrested the cell cycle, and significantly reduced proliferation in breast cancer cells. These findings suggest that miR-335 and -145 may serve as potential therapeutic targets for breast cancer treatment.

Oleanolic Acid's Semisynthetic Derivatives HIMOXOL and Br-HIMOLID Show Proautophagic Potential and Inhibit Migration of HER2-Positive Breast Cancer Cells In Vitro

Natalia Magdalena Lisiak et al.

Summary: This study revealed the cytostatic, proautophagic, and anti-migratory activities of pentacyclic triterpenes' derivatives, specifically HIMOXOL and Br-HIMOLID, in HER2-positive breast cancer cells. Both derivatives showed higher cytotoxicity compared to the maternal compound, OA, and were found to decrease cell viability, induce autophagy, and reduce cell migration in the studied breast cancer cells.
