Association between HSPA8 Gene Variants and Ischemic Stroke: A Pilot Study Providing Additional Evidence for the Role of Heat Shock Proteins in Disease Pathogenesis

Macrophage-Specific NLRC5 Protects From Cardiac Remodeling Through Interaction With HSPA8

Summary: Macrophages play a crucial role in regulating inflammation and tissue repair, particularly in the development of heart failure. The presence of NLRC5 in circulating monocytes and cardiac macrophages was significantly increased in patients with hypertrophic cardiomyopathy. Deletion of NLRC5 in myeloid cells worsened pathological cardiac remodeling and inflammation induced by pressure overload. Mechanistically, NLRC5 interacted with HSPA8 and suppressed the NF-kB pathway in macrophages. The absence of NLRC5 in macrophages led to increased secretion of cytokines like interleukin-6 (IL-6), which had effects on cardiomyocyte hypertrophy and cardiac fibroblast activation. Tocilizumab, an anti-IL-6 receptor antagonist, could be a potential therapeutic strategy for cardiac remodeling and chronic heart failure.

NPC-EVs delivery leads to increased blood concentrations of B and T lymphocytes without affecting cerebral cell counts, suggesting a potential contribution to reverting post-stroke immunosuppression. Furthermore, biodistribution analysis reveals the majority of NPC-EVs to be found in extracranial organs like the liver and lung after systemic administration.
