Beyond PARP1: The Potential of Other Members of the Poly (ADP-Ribose) Polymerase Family in DNA Repair and Cancer Therapeutics

Iain A. Richard et al.

Summary: PARP proteins play diverse roles in cellular functions. PARP1 and PARP2 have been extensively studied in DNA repair and cancer therapeutics. PARPi resistance is a challenge in clinical use. Other members of the PARP family are comparatively understudied in DNA damage repair and tumourigenesis.

Medicinal Chemistry Perspective on Targeting Mono-ADP-Ribosylating PARPs with Small Molecules

Maria Giulia Nizi et al.

Summary: Major advances have been made in defining functions for human mono-ADP-ribosylating PARP enzymes (mono-ARTs), opening up potential applications for treating diseases. The combination of structural biology and medicinal chemistry has led to the development of potent small molecule inhibitors that bind to the catalytic domain. Most of these inhibitors are still in the early stages, but some have shown promising profiles as chemical tools. A compound targeting PARP7 has even progressed to clinical trials.

The Expression Profile of miRNA in Glioma and the Role of miR-339-5p in Glioma

Jie Lin et al.

Summary: This study revealed that there are 49 differentially expressed miRNAs in glioma tissues, and the expression of miR-339-5p is significantly downregulated. MiR-339-5p can regulate the proliferative, apoptotic, invasive, and migratory abilities of glioma U87 cells and might suppress the occurrence and development of glioma.

Genetic factors associated with prostate cancer conversion from active surveillance to treatment

Yu Jiang et al.

Summary: Men diagnosed with low-risk prostate cancer who initially choose active surveillance may eventually convert to active treatment due to genetic factors that predispose to aggressive tumors. This study identified genetic variants associated with conversion from active surveillance to active treatment, suggesting that germline genetics may help individualize management decisions for low-risk prostate cancer.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

SPP1 Promotes Enzalutamide Resistance and Epithelial-Mesenchymal-Transition Activation in Castration-Resistant Prostate Cancer via PI3K/AKT and ERK1/2 Pathways

Xiaocong Pang et al.

Summary: SPP1 plays a crucial role in predicting the development of mCRPC and is closely associated with the malignant progression and enzalutamide resistance in CRPC. Knockdown of SPP1 enhances enzalutamide sensitivity and suppresses invasion and migration of prostate cancer cells. Upregulation of SPP1 is linked to the maintenance of PI3K/AKT and ERK1/2 signaling pathways.
