Cystic kidney disease in tuberous sclerosis complex: current knowledge and unresolved questions

Sebastian Gallo-Bernal et al.

Summary: Tuberous sclerosis complex (TSC) is a genetic disorder caused by mutations in TSC1 or TSC2 genes. Loss of inhibition of cell growth leads to the development of benign tumors, especially kidney cysts. This educational review summarizes current knowledge and emphasizes detection, classification, surveillance, and treatment options for cystic kidney disease in TSC.

Role of glucocorticoid and mineralocorticoid receptors in rainbow trout (Oncorhynchus mykiss) skeletal muscle: A transcriptomic perspective of cortisol action

Jorge E. Aedo et al.

Summary: Cortisol is a crucial regulator of neuroendocrine stress responses in teleost, acting through the modulation of glucocorticoid receptor (GR) and mineralocorticoid receptor (MR). A study using RNA-seq analysis explored the differential contribution of GR and MR in the global transcriptional response in teleost skeletal muscle. The findings highlighted the importance of understanding the role of these receptors in stress response.

Mineralocorticoid Receptor Signaling in the Inflammatory Skeletal Muscle Microenvironments of Muscular Dystrophy and Acute Injury

Zachary M. Howard et al.

Summary: Duchenne muscular dystrophy (DMD) is a muscle degenerative disease caused by the loss of dystrophin protein. Current standard-of-care treatment for DMD has limited efficacy and serious side effects. Mineralocorticoid receptor antagonists (MRAs) have shown promise as a therapeutic target for DMD, as they can improve muscle function and reduce inflammation and fibrosis.

The role of the M-band myomesin proteins in muscle integrity and cardiac disease

Ekaterina P. Lamber et al.

Summary: Transversal structural elements in cross-striated muscles, such as the M-band or the Z-disc, play important roles in anchoring and mechanically stabilizing the sarcomere. Specifically, the M-band acts as a mechanical and signaling hub during contraction and impairment of its function can lead to disease and death. Research on the M-band architecture has focused on the assembly and interactions of three major filamentous proteins, including myomesin-2 and myomesin-3, but limited information is available. Recent studies have linked the myomesin protein family, especially myomesin-2, to serious cardiovascular diseases, highlighting their importance in muscle function and disease.

Aldosterone Inhibits In Vitro Myogenesis by Increasing Intracellular Oxidative Stress via Mineralocorticoid Receptor

Jin Young Lee et al.

Summary: The study showed that aldosterone treatment inhibited muscle differentiation in mouse C2C12 myoblasts in a dose-dependent manner and increased intracellular reactive oxygen species levels. The interaction between mineralocorticoid receptor and aldosterone in myoblasts was confirmed, and blocking this interaction reversed the effects of aldosterone on muscle cells. These findings suggest that aldosterone plays a direct role in muscular deterioration and antioxidant or mineralocorticoid receptor antagonists could be potential therapeutic options for sarcopenia reduction.

Mineralocorticoid receptors in the pathogenesis of insulin resistance and related disorders: from basic studies to clinical disease

Guanghong Jia et al.

Summary: Aldosterone plays a significant role in regulating blood pressure and cardiovascular function by promoting sodium retention and modulating endothelial function. Excessive aldosterone and associated MR activation can impair insulin secretion and metabolic signaling, leading to the development of diabetes and cardiometabolic disorders. Inhibiting MR activation may have beneficial effects in preventing impaired insulin metabolic signaling, type 2 diabetes, and related cardiometabolic disorders.
