Intranasal interferon-beta alleviates anxiety and depressive-like behaviors by modulating microglia polarization in an Alzheimer?s disease model

Mohsen Farhangian et al.

Summary: Patients with Alzheimer's disease (AD) often suffer from neuropsychiatric symptoms (NPS), which greatly impact their quality of life and disease progression. Research suggests that microglial phenotypes play a crucial role in the inflammation associated with AD-related NPS. Interferon-beta (IFN beta) has anti-inflammatory properties and may be an effective treatment for NPS. In this study, IFN beta was administered intranasally to AD rat models and demonstrated to reduce anxious/depressive-like behaviors and promote microglial M2 polarization.

Microglial ERK-NRBP1-CREB-BDNF signaling in sustained antidepressant actions of (R)-ketamine

Wei Yao et al.

Summary: The study revealed that (R)-ketamine exerts its antidepressant-like effects through activation of the ERK-NRBP1-CREB-BDNF pathway in microglia. By increasing the expression of NRBP1, BDNF, and p-CREB/CREB ratio, as well as promoting BDNF transcription and suppressing MeCP2 in microglia, (R)-ketamine demonstrates prolonged antidepressant effects compared to (S)-ketamine. Additionally, microglial depletion or inhibition using specific inhibitors blocked the antidepressant-like effects of (R)-ketamine in susceptible mice, indicating the importance of microglia in mediating the antidepressant actions of (R)-ketamine.

Summary: This review summarizes the research progress on Sigma-1 receptor in the pathogenesis of major depression disorder (MDD), highlighting its involvement in the development of depressive symptoms and its potential as a target for antidepressant development. Agonists of Sigma-1 receptor have been shown to exhibit antidepressant-like activities through various mechanisms.

Neuroinflammation and depression: A review

Romain Troubat et al.

Summary: Neuroinflammation interacts with key neurobiological factors of depression, potentially presenting a crucial therapeutic target for future treatments. This interaction may impact serotonin levels, HPA axis regulation, and hippocampal neurogenesis, leading to the pathophysiological cascade of increased glutamate levels and altered neurotransmission. It can be triggered by chronic inflammatory conditions or primary brain neuroinflammation, highlighting the importance of addressing neuroinflammation in the treatment of major depressive disorder.

Microglia in depression: current perspectives

Xiaoning Jia et al.

Summary: This review focuses on the role of microglia in the etiology of depression, discussing the dynamic states of microglia, the correlative and causal evidence of microglial abnormalities in depression, possible mechanisms of how microglia sense depression-related stress and modulate depression state, and how antidepressive therapies affect microglia. Understanding the role of microglia in depression may lead to new treatment strategies for fighting against this mental illness.

The role of the excitation:inhibition functional balance in the mPFC in the onset of antidepressants

Yong-Yu Yin et al.

Summary: This study demonstrated the effects of ketamine on the E:I functional balance in the rat mPFC and its antidepressant action, while fluoxetine did not significantly impact the E:I balance. Furthermore, chronic treatment with fluoxetine affected the firing activities of pyramidal neurons and interneurons in the mPFC.
