Cancer hallmarks intersect with neuroscience in the tumor microenvironment

Douglas Hanahan et al.

Summary: The multistep process of tumorigenesis can be explained by the acquisition of functional capabilities known as hallmarks of cancer, which are necessary for malignancy. These capabilities exist in transformed cancer cells and the surrounding tumor microenvironment (TME), and are conveyed by abnormal characteristics of the cancerous phenotype. This perspective discusses the role of the nervous system in inducing hallmark capabilities, specifically the involvement of neurons and their projections (axons) in the TME. It also explores the aberrantly activated autocrine and paracrine neuronal regulatory circuits in cancer cells, which contribute to the manifestation of hallmark functions and subsequent cancer pathogenesis.

Potential Toxic Mechanisms of Neonicotinoid Insecticides in Rice: Inhibiting Auxin-Mediated Signal Transduction

Jianjian Wu et al.

Summary: Improper use of pesticides has negative effects on both ecosystem service providers and crop production. This study found that three neonicotinoid insecticides inhibit the auxin signal pathway in rice leaves, leading to a decrease in intracellular auxin content. This disruption in the auxin pathway affects phenylpropanoid biosynthesis and decreases antioxidant metabolites, ultimately reducing rice biomass.

Cancer-cell-derived GABA promotes beta-catenin-mediated tumour growth and immunosuppression

De Huang et al.

Summary: Huang et al. report that cancer cells can rewire glutamine metabolism to synthesize the neurotransmitter GABA, which activates beta-catenin and suppresses T-cell infiltration, promoting tumor growth and leading to resistance to immune checkpoint blockade therapy. Targeting GAD1 or GABA(B)R can overcome this resistance and improve treatment outcomes.

CAMOIP: a web server for comprehensive analysis on multi-omics of immunotherapy in pan-cancer

Anqi Lin et al.

Summary: Immune checkpoint inhibitors (ICIs) and immunotherapy have revolutionized tumor diagnostics and treatment. However, current web tools lack the ability to facilitate data mining and mechanism analyses specifically related to immunotherapy. Therefore, the development of the web-based tool CAMOIP provides a comprehensive analysis of biomarkers for immunotherapy and enables various analyses, promoting the further development of immunotherapy.

Glioblastoma hijacks neuronal mechanisms for brain invasion

Varun Venkataramani et al.

Summary: A subpopulation of glioblastoma cells lacking connections with other tumor cells and astrocytes but receiving synaptic input from neurons drives whole-brain colonization. These cells resemble neuronal and neural-progenitor-like states and invade the brain using mechanisms similar to neuronal migration. Neuronal activity induces calcium signals, leading to the formation of tumor microtubes and increased invasion speed. This study reveals how neuronal mechanisms govern glioblastoma cell invasion and explains the close interlink between tumor dissemination and cellular heterogeneity.

GABA regulates IL-1b production in macrophages

Jian Fu et al.

Summary: The study demonstrates that GABA plays a role in regulating macrophage maturation and inflammation. It enhances certain signaling pathways and reduces the formation of inflammatory complexes, leading to a decrease in pro-inflammatory responses. GABA also shows potential in alleviating sepsis and obesity in mice.

Pioneering insights of extrachromosomal DNA (ecDNA) generation, action and its implications for cancer therapy

Zesheng Li et al.

Summary: This review summarizes the current understanding of the origin, biogenesis process, discovery history, molecular mechanisms, and physiological functions of extrachromosomal DNA (ecDNA) in cancer. Recent studies have revealed that ecDNA plays a crucial role in tumorigenesis and progression, and is associated with poor clinical outcomes and drug resistance. Effective research methods for studying ecDNA and novel insights for ecDNA-directed therapies are also highlighted.

Single-Cell Analysis of the Pan-Cancer Immune Microenvironment and scTIME Portal

Fang Hong et al.

Summary: Single-cell sequencing has provided new insights into the investigation of tumor immune microenvironments (TIME). This study established a pan-cancer single-cell reference of TIMEs, identified new cell type-specific transcription factors, and compared the variation of immune cell types across different patients and tumor types. The study also highlighted the role of tumor-associated macrophages in regulating dysfunctional T cells and the tumor-type specific ligand-receptor communication network within TIMEs.
