Epstein-Barr virus latency programs dynamically sensitize B cells to ferroptosis

Eric M. Burton et al.

Summary: Epstein-Barr virus (EBV) infection plays a role in multiple cancers, and this study examines how EBV infection affects sensitivity to ferroptosis, a form of cell death driven by lipid peroxidation and membrane damage. The study finds that EBV-infected B cells are sensitive to blockage of glutathione biosynthesis and have elevated levels of lipid reactive oxygen species (ROS) that require SLC7A11 and GPX4 activity to prevent ferroptosis. Different stimulation methods affect the sensitivity of B cells to ferroptosis, and EBV-transformed B cells become progressively resistant to ferroptosis induction.

Final Overall Survival Analysis of Gemcitabine and Cisplatin Induction Chemotherapy in Nasopharyngeal Carcinoma: A Multicenter, Randomized Phase III Trial

Yuan Zhang et al.

Summary: This study demonstrated that induction chemotherapy before concurrent chemoradiotherapy significantly improved overall survival in patients with locally advanced nasopharyngeal carcinoma, without increasing the risk of late toxicities. Tumor response to induction chemotherapy and pretreatment cell-free Epstein-Barr virus DNA may serve as useful indicators for individualized treatment.

Augmenting chemotherapy with low-dose decitabine through an immune-independent mechanism

Wade R. Gutierrez et al.

Summary: This study explores the concept of using low-dose Decitabine to enhance the efficacy of chemotherapy. It shows that adding DAC to gemcitabine treatment improves survival and slows tumor growth in a mouse model of high-grade sarcoma. Unlike previous studies, DAC does not induce an antitumor T cell response in sarcoma and the synergy between DAC and gemcitabine is independent of the immune system.

Primary colon natural killer (NK)/T-cell lymphoma, nasal type, with perforations: a case report and literature review

Yuankang Xie et al.

Summary: ENKTL-NT is a rare and aggressive subtype of non-Hodgkin's lymphoma primarily involving the nasal cavity, with rare cases reported in the colon. Early diagnosis is difficult due to the lack of characteristic clinical presentation, leading to frequent misdiagnosis. We presented a case of primary colonic ENKTL-NT in a 51-year-old male patient with abdominal pain as the primary symptom, who showed significant improvement after chemotherapy treatment.

The Role of Lytic Infection for Lymphomagenesis of Human γ-Herpesviruses

Christian Munz

Summary: EBV and KSHV are oncogenic gamma-herpesviruses associated with 1-2% of human tumors, encoding bona fide oncogenes expressed during latent infection. Recent studies have shown that early lytic replication of these viruses supports tumorigenesis, contrary to previous beliefs that lytic infection destroys host cells.

Cedazuridine/decitabine: from preclinical to clinical development in myeloid malignancies

Anand A. Patel et al.

Summary: The development of oral DNA hypomethylating agents has been of interest, with poor oral bioavailability leading to the development of the novel CDA inhibitor cedazuridine. Clinical trials have shown bioequivalence of the oral cedazuridine-decitabine combination in myelodysplastic syndromes and chronic myelomonocytic leukemia.

Methylation alterations and advance of treatment in lymphoma

Meng-Ke Liu et al.

Summary: Lymphoma, a common and aggressive form of hematopoietic malignancies with diverse clinical and pathological features due to its heterogeneity, is influenced by epigenetic alterations, particularly DNA and histone methylation. Targeted therapies such as azacitidine and decitabine have been developed to counteract the pathogenetic consequences induced by aberrant methylation and improve patient outcomes. Investigating and targeting epigenetic mechanisms in lymphoma could be crucial for future research and treatment strategies.
