Rare Variation in Drug Metabolism and Long QT Genes and the Genetic Susceptibility to Acquired Long QT Syndrome

Belinda Gray et al.

Summary: Computational methods and genetic sequencing have identified rare susceptibility variants in CYP genes that are associated with the risk of acquired long QT syndrome (aLQTS). These findings suggest that CYP genes could serve as important markers in personalized medicine approaches in the future.

Pharmacogenetic interventions to improve outcomes in patients with multimorbidity or prescribed polypharmacy: a systematic review

Joseph O'Shea et al.

Summary: Conventional medicines optimisation interventions in people with multimorbidity and polypharmacy are limited, and a more holistic and integrated approach is needed. Pharmacogenetics has the potential to improve medicines optimisation. Current studies suggest that pharmacist-led medicines optimisation, including pharmacogenetics, could have significant benefits for patients and health systems. However, further studies are needed to establish the generalizability of these findings in real-world patient populations.

Whole-genome sequencing of 1,171 elderly admixed individuals from Sao Paulo, Brazil

Michel S. Naslavsky et al.

Summary: This study presents a high-coverage WGS dataset of elderly Brazilians from a mixed population, providing valuable data on non-European and mixed populations. The results demonstrate that WGS can identify previously undescribed variants and genomic segments, as well as alleles from HLA genes. The study also shows the clinical usefulness of reclassifying and validating variants associated with dominantly-inherited Mendelian disorders. Furthermore, the findings highlight the improved potential of whole-genome and HLA imputation using WGS data.

Rare genetic variant burden in DPYD predicts severe fluoropyrimidine-related toxicity risk

Elena De Mattia et al.

Summary: This study demonstrated that the burden of rare DPYD variants is significantly associated with higher risk of severe fluoropyrimidine-related toxicity. Through the use of next generation sequencing and in silico predictive tools, the functional impact of these rare variants was evaluated, providing insights into their potential role in predicting adverse reactions to chemotherapy drugs.

Global distribution of CYP2C19 risk phenotypes affecting safety and effectiveness of medications

Mohitosh Biswas

Summary: This study examined the impact of CYP2C19 genetic variants on the safety and efficacy of clinically important medications, finding that African and European populations are at higher risk of adverse drug responses or toxicities due to specific gene variants. Further large-scale clinical studies are needed to assess the clinical outcomes of these medications in light of CYP2C19 pharmacogenomics effects.

Summary: Pharmacogenetics studies the influence of genetic variation on drug response, finding that the majority of individuals may have atypical responses to at least one drug and on average have atypical responses to 10.3 drugs. Nearly a quarter of participants have been prescribed drugs for which they are predicted to have atypical responses.

Potential of whole-genome sequencing-based pharmacogenetic profiling

Sylvan Manuel Caspar et al.

Summary: Pharmacogenetics plays a key role in personalized medicine, with whole-genome sequencing offering a more comprehensive approach to pharmacogenetic profiling. Short-read next-generation sequencing has limitations, and there may be a shift towards long-read sequencing for improved gene diagnostics and pharmacogenetic profiling in the future.

Comprehensive Allele Genotyping in Critical Pharmacogenes Reduces Residual Clinical Risk in Diverse Populations

Shishi Luo et al.

Summary: Genomic-guided pharmaceutical prescribing is increasingly important, and accurate genotyping of pharmacogenomic genes can be challenging due to their complex genetic architecture. The Helix PGx database provides frequency data for CYP2C9, CYP2C19, CYP2D6, and CYP4F2, demonstrating high diversity in CYP2D6 and higher frequency of rare alleles in non-European populations. Most individuals in the cohort carry high-risk variants in pharmacogenes, suggesting that screening these genes could impact a significant portion of the general population.
