Current possibilities and future opportunities provided by three-dimensional lung ECM-derived hydrogels

Regional and disease specific human lung extracellular matrix composition

Summary: Chronic lung diseases, such as COPD and IPF, exhibit region-specific changes in extracellular matrix (ECM) composition, particularly in collagen and basement-membrane associated proteins. This study compares the ECM composition of whole decellularized lungs and specific anatomical lung regions in non-diseased, COPD, and IPF human lungs. The findings demonstrate distinct ECM signatures in different regions and reveal alterations in ECM composition specific to COPD and IPF, including enrichment of type-III collagen and fibulin in IPF aECM. This study provides methodology for future research and a dataset for identifying novel ECM targets for therapeutics.

An in vitro model of fibrosis using crosslinked native extracellular matrix-derived hydrogels to modulate biomechanics without changing composition

Summary: In fibrotic lung diseases, the composition and structure of the extracellular matrix (ECM) are altered, leading to changes in cellular behavior. This study developed an in vitro model to mimic the altered mechanics of a fibrotic microenvironment. The results showed that crosslinking the ECM-derived hydrogels changed their biomechanical properties and fiber organization, which influenced the differentiation and morphology of fibroblasts.

Summary: Adipose tissue-derived stromal cells (ASCs) are a promising research subject in tissue engineering and regenerative medicine due to their easy acquisition, multipotency, and secretion of factors promoting tissue regeneration. However, their retention in or around lesions after direct administration is poor. To address this issue, ASCs can be "immobilized" in hydrogels such as gelatine methacryloyl (GelMA). This study found that the concentration of GelMA can influence the expression of matrix metalloproteases (MMPs) secreted by ASCs, as well as their morphology and the ultrastructure of the hydrogel.

Bioink based on decellularized porcine lung ECM hydrogels supports 3D culturing of lung-resident MSCs without additional crosslinking, leading to improved cell-matrix interactions and increased expression of key receptors. This approach shows promise for enhancing MSC preconditioning for therapeutic applications.
