The NLRP3 inflammasome modulates tau pathology and neurodegeneration in a tauopathy model

Ilie Cosmin Stancu et al.

Summary: This study reveals the significant role of NLRP3 inflammasome in Alzheimer's disease and related tauopathies, with implications for both tau-associated neurodegeneration and tau propagation.

Central role for p62/SQSTM1 in the elimination of toxic tau species in a mouse model of tauopathy

Maiko Ono et al.

Summary: In this study, evidence is provided that p62 exerts a protective role against neuronal death and neuroinflammation induced by abnormal tau accumulation. By eliminating neurotoxic tau species, p62 may act as a potential therapeutic target for the treatment of tauopathies.

A quantitative in vivo imaging platform for tracking pathological tau depositions and resultant neuronal death in a mouse model

Taeko Kimura et al.

Summary: This study established an in vivo imaging system to quantify tau accumulations and consequent neuronal loss using PET and MRI in a mouse model of neurodegenerative tauopathies. The results showed that the tau accumulations measured by F-18-PM-PBB3 binding were consistent with histopathological findings, and the imaging platform could potentially be used to evaluate candidate anti-tau and neuroprotective drugs.

High-Contrast In Vivo Imaging of Tau Pathologies in Alzheimer's and Non-Alzheimer's Disease Tauopathies

Kenji Tagai et al.

Summary: A panel of radiochemicals for in vivo PET imaging of tau pathologies in AD has been developed, but sensitive detection of FTLD tau inclusions remains challenging. The imaging probe PM-PBB3 successfully captures diverse tau deposits in AD and FTLD, with increased binding observed in diseased patients in a subsequent clinical PET study. Visual inspection of F-18-PM-PBB3-PET images aids in individually based identification of diverse clinical phenotypes of FTLD on a neuropathological basis.

Microglial gene signature reveals loss of homeostatic microglia associated with neurodegeneration of Alzheimer's disease

Akira Sobue et al.

Summary: Analysis of microglia RNA sequencing in mouse models and human brains revealed a significant reduction in homeostatic microglial genes and upregulation of disease-associated microglia genes in neurodegenerative disease models. The reduction of homeostatic microglial genes was correlated with neuronal cell loss in mice, while early AD pathology in humans showed decreased expression of microglia and oligodendrocyte-specific markers.

Microglia become hypofunctional and release metalloproteases and tau seeds when phagocytosing live neurons with P301S tau aggregates

Jack H. Brelstaff et al.

Summary: The study reveals that tau protein aggregation in neurons can trigger inflammation and cellular senescence in microglia, leading to impaired phagocytic function and release of tau aggregates. This process facilitates the spreading of tau protein, indicating a potential mechanism for neurodegenerative diseases.

Distinct microglial response against Alzheimer's amyloid and tau pathologies characterized by P2Y12 receptor

Jun Maeda et al.

Summary: Recent studies have shown a significant down-regulation of microglial P2Y12 receptor levels in neurodegenerative diseases, particularly associated with tau pathology. This finding may serve as a sensitive index for neuroinflammatory responses in Alzheimer's disease-related neurodegenerative processes.
