Changes in white matter microstructure following serial ketamine infusions in treatment resistant depression

Brandon Taraku et al.

Summary: The study found that serial ketamine infusion leads to improvements in white matter neuroplasticity and clinical symptoms in treatment resistant depression (TRD) patients. Ketamine significantly decreases neurite density in occipitotemporal white matter pathways, and the changes in neurite density in tracts connecting the basal ganglia, thalamus, and cortex are associated with improvements in anhedonia in TRD patients.

Summary: The Global Burden of Diseases Study in 2019 showed that mental disorders remained among the top ten leading causes of burden worldwide, with no reduction in burden since 1990. The majority of the burden in disability-adjusted life-years (DALYs) came from years lived with disability (YLDs).

Brain-Derived Neurotrophic Factor Signaling in Depression and Antidepressant Action

Eero Castren et al.

Summary: Neurotrophic factors, especially BDNF, are associated with depression and the action of antidepressant drugs, with impaired BDNF signaling linked to mood disorders. Increased BDNF expression and signaling have been linked to the mechanisms of antidepressant drugs, and BDNF regulates various types of neuronal plasticities in the brain, which are connected to antidepressant action. The hypothesis of a connection between neurotrophic factors and neuronal plasticity with mood disorders and antidepressant action has been further supported by recent evidence.

Functional connectivity between the amygdala and subgenual cingulate gyrus predicts the antidepressant effects of ketamine in patients with treatment-resistant depression

Tomoyuki Nakamura et al.

Summary: This study demonstrated the clinical utility of resting-state functional magnetic resonance imaging in predicting the antidepressant effects of ketamine in treatment-resistant depression patients. Significant baseline resting-state functional connectivity differences between ketamine responders and nonresponders were identified, allowing for the prediction of treatment response with high sensitivity and specificity. This suggests that resting-state functional connectivity alterations may play a role in determining the trait-like pathophysiology underlying treatment response heterogeneity in treatment-resistant depression.
