The Epigenetics of Psychosis: A Structured Review with Representative Loci

This review focuses on three genes, MCHR1, AKT1, and TDO2, as models for differential methylation, and explores the environmental factors associated with psychotic disorders and their interaction with these genes. The location of transcription factor motifs relative to key methylation sites is evaluated and evidence is presented for methylation directing alternative splicing. Experimental results from key studies show differential methylation patterns in psychosis cases versus controls, the influence of pre-existing methylation patterns on brain activation following acute administration of psychosis-eliciting environmental stimuli, and a pattern of methylation associated with developmental risk factors for psychosis.

Genome-wide analysis of schizophrenia and multiple sclerosis identifies shared genomic loci with mixed direction of effects

Summary: Despite a negligible genetic correlation, there is polygenic overlap between schizophrenia and multiple sclerosis, with substantial genetic enrichment in each disorder conditional on associations with the other. Leveraging this cross-disorder enrichment, 36 loci jointly associated with schizophrenia and multiple sclerosis were identified at conjunctional FDR < 0.05 with mixed direction of effects. By conducting functional analysis, candidate genes and biological processes related to immune response and B-cell receptor signaling pathways were implicated in the shared genetic architecture of these two disorders.
