Upregulation of the ERR & gamma;-VDAC1 axis underlies the molecular pathogenesis of pancreatitis

Dipanjan Chanda et al.

Summary: Emerging evidence suggests that the transcription factor ERRα plays a vital role in the development of pancreatitis. Pancreatitis is associated with an increase in ERRα gene expression, and its inhibition impairs the progression of the disease. Mechanistically, ERRα induces the expression of VDAC1, which modulates mitochondrial Ca2+ and ROS levels. Inhibition of the ERRα-VDAC1 axis alleviates mitochondrial dysfunction and suppresses pancreatitis progression. The therapeutic blockade of ERRα-VDAC1 pathway shows promise in mitigating pancreatitis in mouse models and human hereditary pancreatitis. These findings emphasize the importance of ERRα in pancreatitis and suggest its potential as a therapeutic target.

Advances in acute and chronic pancreatitis

Williamson B. Strum et al.

Summary: This article reviews articles published between 2019-2022 from international sources and identifies four crucial new findings in acute pancreatitis and chronic pancreatitis. These findings include new genetic pathogenic mutations, a new serum diagnostic marker for AP, explanations of the impact of monocytes/macrophages, and authoritative recommendations for medical treatment of chronic pain. These advances provide opportunities to enhance outcomes, alter the natural history, and envision the future of these diseases.

Emerging ultrasmall luminescent nanoprobes for in vivo bioimaging

Shihua Li et al.

Summary: Photoluminescence (PL) imaging is a critical tool for disease diagnosis, therapeutic evaluation, and surgical navigation. However, developing high-efficiency nanoprobes for in vivo imaging and clinical translation remains a challenge. This review discusses the progress in engineering sub-10 nm ultrasmall luminescent nanoprobes for in vivo PL bioimaging and highlights their unique advantages. The challenges in this field and strategies for improving in vivo PL bioimaging and clinical applications are also addressed.

FRET as the tool for in vivo nanomedicine tracking

Norraseth Kaeokhamloed et al.

Summary: Advanced drug delivery system using nanocarriers is a major application of nanotechnology in pharmacotherapeutics. However, the clinical translation of nanomedicine faces challenges, and this review focuses on utilizing Fo•rster resonance energy transfer (FRET) as a method to track intact nanocarriers in vivo for studying pharmacokinetics.

The Pancreas and Known Factors of Acute Pancreatitis

Julia Walkowska et al.

Summary: Pancreatitis is considered one of the most complicated and clinically challenging disorders affecting the abdomen. The main causes of acute, recurrent acute, and chronic pancreatitis are gallstone migration and alcohol abuse. This study aims to present the known risk factors for acute pancreatitis, as well as the morphology, physiology, and development of the pancreas.

Ruthenium red attenuates acute pancreatitis by inhibiting MCU and improving mitochondrial function

Xiuyan Yu et al.

Summary: This study suggests that the MCU inhibitor, Ruthenium red (RuR), has therapeutic potential in acute pancreatitis (AP). The findings demonstrate that RuR can alleviate AP by reducing pathological damage and inflammation in a mouse model, as well as preventing cell necrosis by maintaining mitochondrial function at the cellular level.

Multi-enzyme mimetic ultrasmall iridium nanozymes as reactive oxygen/ nitrogen species scavengers for acute kidney injury management

Dong-Yang Zhang et al.

Summary: This study introduced ultrasmall polyvinylpyrrolidone-coated iridium nanoparticles as a promising new artificial enzyme to scavenge RONS and protect against AKI. The nanoparticles showed preferential renal uptake and alleviated clinical symptoms in mice, with relatively low systemic side effects due to rapid renal clearance. The research demonstrates the clinically translatable potential of ultrasmall nanozymes for AKI management.

The role of imaging in acute pancreatitis

Maria Gabriella Brizi et al.

Summary: Acute pancreatitis, ranging from mild to severe forms with significant complications, is commonly diagnosed and evaluated using ultrasound, CT, and MRI. CT is the most frequently performed imaging test, while MRI is advantageous in specific situations for better soft tissue contrast resolution and evaluation of ductal disconnection. The revised Atlanta classification system aids radiologists in writing accurate reports and highlights their crucial role in the multidisciplinary team for treating patients with acute pancreatitis.

Prussian blue nanozyme-mediated nanoscavenger ameliorates acute pancreatitis via inhibiting TLRs/NF-κB signaling pathway

Xue Xie et al.

Summary: The study found that PBzyme has potential anti-oxidative and anti-inflammatory effects in the prophylactic treatment of AP, possibly involving inhibition of signaling pathways and scavenging reactive oxygen species. This provides a foundation for the clinical application of PBzyme in the preventive treatment of AP.
