Heritable transgene-free genome editing in plants by grafting of wild-type shoots to transgenic donor rootstocks

Summary: Generation of transgene-free plant lines in one generation without the need for transgene elimination, culture recovery and selection, or use of viral editing vectors has been achieved by designing fusions of Cas9 and guide RNA transcripts to tRNA-like sequence motifs that facilitate RNA movement between transgenic rootstocks and wild-type scions. The graft-mobile gene editing system has been successfully demonstrated in Arabidopsis thaliana and Brassica rapa, and may have broad applications in breeding programs and crop plants.

Small RNA populations reflect the complex dialogue established between heterograft partners in grapevine

Summary: In this study, the populations of small RNAs in homo and heterografts were analyzed, and the possible exchange of small RNAs between heterograft partners was explored by taking advantage of sequence differences in their genomes. It was found that the type of grafting and genotype have significant effects on small RNA populations. Additionally, bilateral exchanges of small RNAs, especially short interfering RNAs, were observed in heterografts with a preference for transfer from scions to rootstocks. Overall, these findings suggest that small RNAs may play an important role in the phenotype modifications observed in heterografts.

Arabidopsis apoplastic fluid contains sRNA- and circular RNA-protein complexes that are located outside extracellular vesicles

Summary: By studying the apoplastic wash fluid of Arabidopsis leaves, it was found that small RNAs and long noncoding RNAs, including circular RNAs, are mainly located outside the extracellular vesicles and are associated with protein complexes. These extracellular RNAs are highly enriched in the posttranscriptional modification N-6-methyladenine (m(6)A). The study suggests that these extracellular RNAs may play a role in host-induced gene silencing.
