A combined approach of DNA metabarcoding collectively enhances the detection efficiency of medicinal plants in single and polyherbal formulations

Detection of Carica papaya Adulteration in Piper nigrum Using Chloroplast DNA Marker-Based PCR Assays

Summary: This study developed a PCR assay for detecting the adulteration of Carica papaya (CP) seeds in Piper nigrum (PN) products. Through the design of specific primers and optimization of PCR conditions, the target DNA sequence of CP seeds could be amplified down to 0.1 ng. Additionally, simplex, duplex, and digital PCR (dPCR) assays were able to detect 1.0% (w/w) adulteration of CP seeds in blended formulations.

A duplex PCR assay for authentication of Ocimum basilicum L. and Ocimum tenuiflorum L in Tulsi churna

Summary: This study developed a simple and cost effective PCR method, as well as a duplex PCR assay, for the identification of Ocimum basilicum and Ocimum tenuiflorum. The methods were validated using molecular and chromatographic authentication methods on market samples, and the combination of both methods was found to achieve maximum fidelity.

Decoding herbal materials of TCM preparations with the multi-barcode sequencing approach

Summary: With the development of high-throughput sequencing technology, the multi-barcode sequencing approach has been successfully used to identify biological ingredients in Traditional Chinese Medicine (TCM) preparations. In this study, the approach was applied to four representative TCM preparations, and the results showed good universality, sensitivity, and reliability in decoding the herbal materials in these preparations.

Strategies for molecular authentication of herbal products: from experimental design to data analysis

Summary: Molecular herbal authentication has become popular worldwide in the past decade. DNA-based methods, such as DNA barcoding and species-specific amplification, have been widely adopted for herbal identification. The development of next-generation sequencing (NGS) has significantly increased the throughput of sequencing and facilitated the collection and assembly of organelle genomes. NGS allows simultaneous sequencing of multiple reads, enabling the identification of multiple species from a single sample. This review provides an introduction to the use of DNA barcoding and genome skimming/shotgun metagenomics in herbal product authentication, and discusses important considerations in experimental design for NGS-based botanical identification.
