Virus-Like Particles: Revolutionary Platforms for Developing Vaccines Against Emerging Infectious Diseases

Summary: Virus-like particles (VLPs) are nanostructures with various applications in therapeutics, immunization, and diagnostics. Recent advancements in biomedical engineering have made commercially available VLP-based vaccines widely used in combating infectious diseases. However, there are production and fabrication challenges that need to be addressed before VLP-based approaches can be widely used. This review provides insights into the characteristics, expression systems, and potential applicability of VLP-based vaccines, emphasizing their potential as ideal candidates to combat emerging virulent pathogens and induce immunity against them.

The generation of empty Turnip yellow mosaic virus capsids through depletion of virion-associated divalent cations

Summary: This study presents a convenient method for generating empty particles from Turnip yellow mosaic virus (TYMV) through EDTA treatment. The findings reveal that TYMV virions purified in an EDTA-based buffer are converted to 94% empty particles on average. Additionally, the purification method used for TYMV virions influences their conversion to empty particles after freeze-thaw.

The impact of size on particle drainage dynamics and antibody response

Summary: In this study, engineered CCMVTT-VLPs were investigated for their draining kinetics and immunogenicity, with Round-shaped CCMVTT-VLPs showing more efficient drainage to secondary lymphoid organs and greater systemic IgG and IgA responses compared to Rod-shaped CCMVTT-VLPs. Additionally, Round-shaped CCMVTT-VLPs led to prominent formation of splenic germinal centers and polarization of T cell response towards Th1. This is the first study to compare the draining kinetics and immunogenicity of VLPs forming nano-sized icosahedra or micrometer-sized rods.
