Quercetin derivatives: Drug design, development, and biological activities, a review

Seyedeh Roya Alizadeh et al.

Summary: More studies are needed to develop new drugs that can overcome drug resistance and minimize side effects. Quercetin, a natural flavonoid, has shown diverse biological activities, but its poor solubility and bioavailability limit its applicability. Researchers have been exploring the modification of quercetin by designing and synthesizing novel derivatives to overcome these limitations. The physico-chemical properties of quercetin's molecular scaffold make it attractive for drug development, and the investigation of its derivatives' activities, chemical structure-activity relationship, and mechanism of action is crucial. Quercetin-based molecules have the potential to contribute to the discovery of medications for various diseases.

Summary: This study investigated the antiviral effects of GS-9620 against foot-and-mouth disease virus (FMDV). It was found that GS-9620 inhibited the virus in swine cells and induced the production of interferons and cytokines, providing early protection. Furthermore, the combination of GS-9620 with an inactivated vaccine was highly effective.

Natural Phytochemicals, Luteolin and Isoginkgetin, Inhibit 3C Protease and Infection of FMDV, In Silico and In Vitro

Sirin Theerawatanasirikul et al.

Summary: By utilizing computer-aided virtual screening of natural phytochemical compound libraries, the study identified luteolin and isoginkgetin as promising antiviral agents for FMDV. These compounds exhibited significant antiviral effects, particularly isoginkgetin which was found to be a potent inhibitor of FMDV 3C(pro). Further analysis suggested that both compounds interacted effectively with key enzymatic residues of the 3C(pro), indicating their potential for development as antiviral drugs against FMDV and other picornaviruses.

Use of Protein Pegylation to Prolong the Antiviral Effect of IFN Against FMDV

Fayna Diaz-San Segundo et al.

Summary: Interferons are considered as the first line of defense against viral diseases, and they have become an attractive therapeutic option to control virus infections. In this study, pegylated recombinant porcine interferon alpha showed strong and long-lasting antiviral activity against foot-and-mouth disease virus in vitro and in vivo, providing complete protection against the disease for at least five days after a single dose. These results highlight the potential of this biotherapeutics to be used in combination with vaccines for full control of foot-and-mouth disease in the field.
