Exploration of Potent Cytotoxic Molecules from Fungi in Recent Past to Discover Plausible Anticancer Scaffolds

Summary: Fungi can produce various scaffolds with unique biological activities, but so far, no molecule derived from fungi has become an anti-cancer drug on the market. Every year, numerous cytotoxic molecules of fungal origin are published, making it necessary to critically analyze these compounds to identify the potent ones. A review published in 2014 mentioned the best cytotoxic fungal metabolites and their status in drug development. In this report, we included 176 cytotoxic molecules isolated from fungi after 2014 and categorized them based on their potencies. Emphasis was given to discuss the 42 molecules that showed IC50 values below 1 mu M. This review provides potent fungal scaffolds that can be prioritized as drug candidates for cancer therapeutics.

Molecular Networking-Based Screening Led to the Discovery of a Cyclic Heptadepsipeptide from an Endolichenic Xylaria sp

Summary: This study utilized MS/MS-based molecular networking strain prioritization to discover a group of cyclic depsipeptides from an endolichenic Xylaria sp. The main component, xylaroamide A (1), was isolated using LC-MS guidance. Elucidation of the compound's structure and configuration was achieved through ID and 2D NMR, as well as MS/MS data. The compound exhibited inhibitory activity against cancer cell lines BT-549 and RKO, making it a potentially valuable candidate for drug development.

Assessing Microbial Metabolic and Biological Diversity to Inform Natural Product Library Assembly

Summary: The need for new chemical matter to support the discovery of bioactive compounds has led natural product researchers to explore various source organisms and environments. This study compared fungi from different biomes and found modest differences in physiological performance and chemical output, but no significant differences in chemical richness. Certain fungal genera were more strongly associated with aquatic sediments compared to the surrounding terrestrial environment. These findings suggest that focusing efforts on sampling unique microbial resources in an environment may enhance the diversity of natural products needed for chemical discovery.

Progress Report: Antimicrobial Drug Discovery in the Resistance Era

Summary: Antibiotic resistance poses a serious threat to public health, and alternative strategies are needed. The GLASS report reveals an increasing number of bacterial infections caused by antimicrobial-resistant bacteria, leading to educational efforts on the appropriate use of antibiotics by government agencies and organizations. This review focuses on the historical development of antimicrobial drug discovery, discusses new targets and approaches, advancements in drug screening, design, and delivery, as well as the emergence of antibiotic adjuvants and combination therapies. Despite the challenges posed by resistant strains, alternative technologies such as nanomaterials show promise for the future.

7-Hydroxy-2-octenoic acid-ethyl ester mixture as an UV protectant secondary metabolite of an endolichenic fungus isolated from Menegazzia terebrata

Summary: In this study, a partially purified compound, 7-hydroxy-2-octenoic acid-ethyl ester (7E), was obtained from the secondary metabolites of enodolichenic fungi (ELF). The compound showed potential in UV absorption and antioxidant properties, with the ability to repair damage caused by UV radiation and inhibit melanin synthesis.

Antibacterial Effect and Mode of Action of Secondary Metabolites from Fungal Endophyte Associated with Aloe ferox Mill

Summary: This study assessed the antibacterial effect and mode of action of secondary metabolites from fungal endophyte associated with Aloe ferox Mill. One particular fungus, Aspergillus welwitschiae MK450668.1, showed promising antibiotic production and inhibited the activity of respiratory chain dehydrogenase and membrane integrity in the target bacteria. These findings suggest that A. welwitschiae MK450668.1 may serve as a potential source of effective compounds to combat drug resistance.

A new alkanol from the endolichenic fungus Daldinia childiae

Summary: One new alkanol, 2,4,5-heptanetriol(1), along with four compounds, was isolated from the ethyl acetate extract of rice solid-substrate cultures of endolichenic fungus Daldinia childiae for the first time. Compound 3 showed alpha-amylase inhibitory activity, marking the first separation of compounds 1, 2, and 3 from the genus Daldinia.

Alkylated Salicylaldehydes and Prenylated Indole Alkaloids from the Endolichenic Fungus Aspergillus chevalieri and Their Bioactivities

Summary: Sixteen metabolites, including seven C-7-alkylated salicylaldehyde derivatives and nine prenylated indole alkaloids, were isolated from the endolichenic fungus Aspergillus chevalieri SQ-8. Some of these compounds exhibited potent antibacterial, antineuroinflammatory, and antioxidant activities, with asperglaucins A and B showing potential as lead compounds for agrochemical bactericides. Additionally, compounds 6 and 10 demonstrated significant inhibition of nitric oxide production and phenolics 3-5 displayed antioxidative effects, suggesting the strain SQ-8 as a novel resource for bioactive metabolites.
