Recent advances on dual inhibitors targeting HIV reverse transcriptase associated polymerase and ribonuclease H

Summary: Reverse transcriptase (RT) is crucial for the replication of HIV and dual inhibitors targeting HIV RT associated polymerase and RNase H have been developed to overcome drug resistance. These dual inhibitors have advantages in increasing efficacy, reducing toxicity, and improving patient compliance. This review summarizes recent advances in drug design strategies and structure-activity relationships of polymerase/RNase H dual inhibitors, providing new insights into the development of anti-HIV drugs.

HIV-1 3′-Polypurine Tract Mutations Confer Dolutegravir Resistance by Switching to an Integration-Independent Replication Mechanism via 1-LTR Circles

Summary: Recent studies have shown that mutations in the HIV-1 3' polypurine tract (3' PPT) motif can reduce sensitivity to the integrase inhibitor dolutegravir (DTG). Multiple mutations in this viral RNA element confer DTG resistance by inactivating a critical reverse transcription element. These mutant viruses replicate without integration into the host cell genome, suggesting a novel escape mechanism from DTG.

Novel RNase H Inhibitors Blocking RNA-directed Strand Displacement DNA Synthesis by HIV-1 Reverse Transcriptase

Summary: In retroviruses, strand displacement DNA-dependent DNA polymerization catalyzed by viral reverse transcriptase (RT) is crucial for synthesizing double-stranded proviral DNA. This study demonstrates that the loss of RNase H activity in HIV-1 RT does not significantly affect strand displacement during DNA template copying but has a major impact on DNA polymerization during reactions with RNA templates. The findings suggest potential applications in transcriptomics technologies and the development of novel antiretroviral agents.

Mutations in the 3'-PPT Lead to HIV-1 Replication without Integration

This mutation altered the reverse transcription process, leading to the accumulation of 1-LTR circular structures instead of linear DNA.
