The Hha-TomB toxin-antitoxin module in Salmonella enterica serovar Typhimurium limits its intracellular survival profile and regulates host immune response

Prajita Paul et al.

Summary: This study investigated the role of the Hha-TomB TA system in regulating the virulence of Salmonella enterica serovar Typhimurium, revealing that deletion of hha and tomB genes leads to impaired cell adhesion, invasion, and uptake, as well as reduced intracellular replication. The TA mutants did not induce acute colitis in mice and showed decreased dissemination to systemic organs. Additionally, inflammation phenotype was restored upon complementation of the TA genes.

Whole Genome Sequencing, Antibiotic Resistance, and Epidemiology Features of Nontyphoidal Salmonella Isolated From Diarrheic Children: Evidence From North China

Wei Zhao et al.

Summary: This study characterized 260 strains of human Salmonella isolated from Jilin, China through serotyping and antimicrobial resistance analysis using whole genome sequencing. The most common serotype was Salmonella enteritidis, followed by S. I 4,[5],12:i:- and Salmonella Typhimurium. The presence of resistance genes and their corresponding phenotypes were confirmed, as well as the presence of specific mutations related to quinolone resistance. The combination of multilocus sequence typing (MLST) and core genome multilocus sequence typing (cgMLST) proved to be effective in molecular traceability. Comprehensive surveillance of Salmonella infections in children was recommended to monitor antimicrobial-resistant trends and prevent outbreaks of foodborne diseases, thus protecting public health.

Evaluation of the Protective Immune Response Induced by an rfbG-Deficient Salmonella enterica Serovar Enteritidis Strain as a Live Attenuated DIVA (Differentiation of Infected and Vaccinated Animals) Vaccine in Chickens

Xinwei Wang et al.

Summary: In this study, a genetically modified Salmonella vaccine was constructed and its protective effectiveness and DIVA potential in chickens were evaluated. Vaccinated chickens showed a significant humoral immune response and no clinical symptoms, pathological changes, or death after challenge.

Activation of apoptosis by Salmonella pathogenicity island-1 effectors through both intrinsic and extrinsic pathways in Salmonella-infected macrophages

Hsin-Hung Lin et al.

Summary: The study revealed that Salmonella induces apoptosis in macrophages through caspase-3 activation, especially via the expression of SPI-1 structure proteins and effectors. This mechanism may help explain why Salmonella infections in children often present as mild bacteremia with less systemic inflammatory response.

Salmonella Typhimurium reprograms macrophage metabolism via T3SS effector SopE2 to promote intracellular replication and virulence

Lingyan Jiang et al.

Summary: S. Typhimurium infection induces upregulated glycolysis and decreased serine synthesis in host macrophages, leading to accumulation of glycolytic intermediates that promote bacterial replication and virulence.

Salmonella Typhimurium impairs glycolysis-mediated acidification of phagosomes to evade macrophage defense

Saray Gutierrez et al.

Summary: This study reveals how Salmonella Typhimurium disrupts macrophage defense by abrogating glycolysis and its regulators, leading to impaired bacterial clearance and antigen presentation. Additionally, glycolysis is shown to facilitate v-ATPase assembly and phagosome acidification, crucial for lysosomal degradation in macrophages. The findings highlight the molecular link between metabolic adaptation and phagosome maturation, targeted by S. Typhimurium to evade host defense mechanisms.
