Vision-dependent specification of cell types and function in the developing cortex

Sarah Cheng et al.

Summary: This study explores the role of postnatal experience in shaping cortical circuitry at the level of cell types in the mouse primary visual cortex. The findings suggest that vision selectively drives the development of upper-layer glutamatergic cell types, while deeper-layer cell types are established prior to eye opening. Furthermore, vision also influences the formation of a spatial continuum of L2/3 cell types through the regulation of specific gene expression programs. One of these vision-dependent genes, Igsf9b, is crucial for the normal development of binocular responses in L2/3.

Rectifying and sluggish: Outer hair cells as regulators rather than amplifiers

Marcel van der Heijden et al.

Summary: This study highlights the mechano-electrical transduction and dynamic range compression in the cochlea, focusing on the electromotility of outer hair cells. Experimental findings suggest that high-frequency tones can affect the receptor potential of OHCs and mechanical amplification may have adverse effects on audibility, while cochlear traveling waves are efficient in transporting high-frequency energy with minimal losses. The study also reveals the role of OHC motility in regulating local dissipation processes.

Opioid receptor activation modulates the calcium current in the cochlear outer hair cells of the rat

Rosario Vega et al.

Summary: This study investigated the modulation of Ca2+ currents in rat outer hair cells by opioid peptides. The results showed that activation of kappa opioid receptors inhibits Ca2+ currents, affecting intracellular Ca2+ concentration. This suggests that opioids have a significant role in the modulation of auditory function and may contribute to the development of auditory disturbances.

What is a cell type and how to define it?

Hongkui Zeng

Summary: Cell types are the fundamental units of an organism's function and exhibit diverse phenotypic properties. This review provides an overview of the principles of cell types rooted in evolution and development, focusing on the mammalian brain. It discusses approaches to characterize and classify cell types and proposes a roadmap for understanding the dynamic changes in cellular function under different conditions.

Electrical signaling in cochlear efferents is driven by an intrinsic neuronal oscillator

Hui Hong et al.

Summary: Efferent neurons, specifically the lateral olivocochlear (LOC) neurons, play vital roles in maintaining auditory function. Through in vitro experiments, it was discovered that LOC neurons in juvenile and young adult mice exhibited extremely slow waves of activity driven by an intrinsic oscillator dependent on L-type Ca2+ channels. Additionally, optogenetic approaches helped identify both ascending and descending sources of synaptic excitation, as well as the synaptic receptors involved. These findings provide important insights into the mechanisms of diverse transmitter release by LOC fibers in the cochlea.

Electron Microscopic Reconstruction of Neural Circuitry in the Cochlea

Yunfeng Hua et al.

Summary: Recent studies have shown a great diversity in the afferent synaptic connections between IHCs and SGNs, with varying abundance and composition of efferent innervation depending on the connectivity of the afferent SGN dendrites with IHCs. Different levels of efferent innervation are provided to SGNs based on the number and source of stimuli received, and branched SGN dendrites and medial olivocochlear neurons were also found to play a role in efferent innervation of SGN dendrites.

Morphological diversity of single neurons in molecularly defined cell types

Hanchuan Peng et al.

Summary: The study systematically examined the complete single-neuron morphologies on a brain-wide scale and identified 11 major projection neuron types with distinct features. The research found extensive projectional diversity within each major type, following generalizable principles governing long-range axonal projections including molecular correspondence, divergent or convergent projection, axon termination pattern, and regional specificity. This study highlights the crucial need for quantitative description of complete single-cell anatomy in cell-type classification.

Effects of Calcitonin-Gene-Related-Peptide on Auditory Nerve Activity

Colleen G. Le Prell et al.

Summary: The study found that endogenous CGRP released by lateral olivocochlear neurons can enhance auditory nerve activity, but the CGRP receptor antagonist does not affect spontaneous or sound-driven auditory nerve activity or OHC function. The results suggest that CGRP acts on neurons rather than outer hair cells.

Sensory neuron-derived TAFA4 promotes macrophage tissue repair functions

Guillaume Hoeffel et al.

Summary: This study investigated the role of sensory neurons in promoting the tissue-repair function of macrophages and uncovered the regulatory mechanism of the neuropeptide TAFA4 on macrophage inflammatory responses. TAFA4, produced by a subset of sensory neurons, modulates the inflammatory profile of macrophages and promotes the production of IL-10 after skin damage in mice. This neuroimmune regulatory pathway driven by TAFA4 helps prevent fibrosis and promotes tissue regeneration, presenting new therapeutic perspectives for inflammatory diseases.

Hypothalamic dopamine neurons motivate mating through persistent cAMP signalling

Stephen X. Zhang et al.

Summary: The study reveals that mating drive and persistence in male mice are controlled by dopamine neurons. Exposure to females triggers interest for a short time, while successful mating leads to satiety. Accumulation or suppression of dopamine signals regulates mating behaviors across minutes and days.
