Genetic analysis of the human microglial transcriptome across brain regions, aging and disease pathologies

Genetic Analysis of Lung Cancer and the Germline Impact on Somatic Mutation Burden

Summary: This study identified 8 novel LC susceptibility loci through GWAx-GWAS meta-analysis, involving DNA repair genes, metabolic genes, and smoking propensity genes. Polygenic risk score analysis demonstrated the contribution of these variants to LC genetic risk prediction. Patients with higher genetic PRS loads of smoking-related variants tended to have higher mutation burdens in their lung tumors.

Genetic diversity fuels gene discovery for tobacco and alcohol use

Summary: This study utilized global genetic diversity data to discover and fine-map genomic loci associated with tobacco and alcohol use. Ancestry-aware transcriptome-wide association studies were conducted to understand the function of these loci. The study also evaluated the genetic architecture and predictive power of polygenic risk in different populations. The results demonstrated that increasing sample size and genetic diversity improved the identification and fine-mapping of loci, but polygenic risk scores performed poorly across different ancestries.

Polygenic basis and biomedical consequences of telomere length variation

Summary: This study characterized the genetic architecture of leukocyte telomere length in UK Biobank participants and identified 197 sentinel variants associated with LTL at 138 genomic loci. Genetically determined differences in LTL were found to be associated with various biological traits and diseases, and individuals with shorter LTL had a lower life expectancy compared to those with longer LTL. The findings provide insights into the genetic regulation of LTL, as well as its impact on physiological traits, diseases, and longevity.

The authors propose HyPrColoc, an efficient Bayesian clustering algorithm, to detect colocalized traits from large numbers of traits.
