Clonal Expansion of Stem/Progenitor Cells in Cancer, Fibrotic Diseases, and Atherosclerosis, and CD47 Protection of Pathogenic Cells

R. Majeti et al.

Summary: We have proposed and demonstrated the existence of a preleukemic phase in myelogenous leukemia, where normal hematopoietic stem cells gradually accumulate mutations and expand, resulting in the emergence of leukemic stem cells. This phase is not only associated with clonal hematopoiesis but also late-onset blood diseases such as chronic-phase chronic myeloid leukemia, myeloproliferative neoplasms, and myelodysplastic disease.

Multistage feedback-driven compartmental dynamics of hematopoiesis

Nathaniel Vincent Mon Pere et al.

Summary: Human hematopoiesis shows resilience to disruptions, but certain disorders can push the system beyond its natural adaptability, resulting in abnormal blood circulation. Studies have found that a stable response to perturbations requires simultaneous adaptation of cell differentiation and self-renewal rates, and under conditions of continuous disruption, compartment cell numbers evolve to new stable states.

Mathematical modelling of the hematopoietic stem cell-niche system: Clonal dominance based on stem cell fitness

Rasmus Kristoffer Pedersen et al.

Summary: This study presents and analyzes a mechanism-based mathematical model to investigate the interaction between HSC and the stem cell niche. The importance of HSC self-renewal and their role in long-term maintenance of blood cell production is revealed. Two reduced models of different complexity are proposed, one of which aligns well with clinical data and offers a potential prediction of patient prognosis. The mathematical model and reduced forms have the potential to provide insights into the regulation of HSC dynamics and blood cell formation, advancing the treatment of hematologic malignancies in the future.

A Quantitative Paradigm for Decision-Making in Precision Oncology

Dalit Engelhardt et al.

Summary: The study proposes a quantitative paradigm for cancer therapeutic decision-making that is dynamic, personalized, and capable of identifying optimal treatment strategies under uncertainty. Comprehensive integration of clinical and molecular data is crucial, and mathematical modeling and machine learning play key roles in constructing dynamic optimal treatment strategies. Reinforcement learning approaches show potential in this endeavor.

Optimal experimental design for mathematical models of haematopoiesis

Luis Martinez Lomeli et al.

Summary: This study aims to uncover the mechanisms in haematopoiesis through perturbation experiments, using a novel Bayesian hierarchical framework for optimal experimental design and data analysis. The research utilizes a deterministic model and principles of Bayesian experimental design to overcome the obstacle of non-longitudinal experimental data, showing that optimal design can lead to better parameter estimates.
