Construction of engineered RuBisCO Kluyveromyces marxianus for a dual microbial bioethanol production system

Dung Minh Ha-Tran et al.

Summary: This study successfully transformed RubisCO genes from Rhodopseudomonas palustris into Kluyveromyces marxianus, and utilized Hungateiclostridium thermocellum for efficient solubilization of lignocellulosic biomass to generate fermentable sugars. The engineered K. marxianus strains showed efficient bioethanol production from Napier grass and rice straw, indicating the potential of a bacterium-yeast coculture system for ethanol production directly from biomass feedstocks.

Auxiliary Module Promotes the Synthesis of Carboxysomes in E. coli to Achieve High-Efficiency CO2 Assimilation

Yuwei Zhang et al.

Summary: A study designed an auxiliary module to help E. coli self-assemble into cyanobacterial CBs, improving the structural integrity and CO2-concentrating ability of the synthetic CBs. The engineered strain efficiently converted external CO2 to metabolic intermediates in the central metabolic pathway, showing potential for understanding CO2 assimilation mechanisms and applications in synthetic biology.

Discovery of a readily heterologously expressed Rubisco from the deep sea with potential for CO2 capture

Junli Zhang et al.

Summary: The Form II Rubisco from Riftia pachyptila endosymbiont, RPE Rubisco, shows a significantly higher carboxylation efficiency compared to other Rubisco enzymes, making it an ideal enzyme for CO2 capture without the need for complex co-expression of chaperones. Additionally, the CO2-fixing endosymbionts may have greater efficiency than previously realized, with potential for future microbial CO2 sequestration platforms.

Stepwise optimization of genetic RuBisCO-equipped Escherichia coli for low carbon-footprint protein and chemical production

Shih- Tan et al.

Summary: The study focuses on enhancing CO2 assimilation capability in E. coli by introducing RuBisCO and PRK genes along with the CRISPRi system. This led to improved efficiency in simultaneous CO2 recycling and production of high-value molecules. The genetically engineered RuBisCO-equipped E. coli has the potential to be a low-carbon microbial cell factory with significant environmental benefits.
