Rab11 endosomes and Pericentrin coordinate centrosome movement during pre-abscission in vivo

Nikhila Krishnan et al.

Summary: The last stage of cell division involves the cleavage of a cytokinetic bridge that connects two daughter cells. Rab11-endosomes are organized in a Rab11-GTP dependent manner at the mother centriole during pre-abscission, with the oldest centrosome showing more mobile Rab11-endosomes compared with the youngest. Rab11-endosomes contribute to centrosome function during pre-abscission by regulating Pericentrin organization, resulting in appropriate centrosome movement towards the cytokinetic bridge and subsequent abscission.

Pericentriolar matrix (PCM) integrity relies on cenexin and polo-like kinase (PLK)1

Abrar Aljiboury et al.

Summary: The study indicates that cenexin regulates microtubule nucleation by maintaining PCM cohesion in a PLK1-dependent manner. Wild-type human cenexin can rescue the PCM architecture in cenexin-depleted zebrafish embryos. The C terminus of cenexin acts in a conserved manner in eukaryotes, excluding nematodes and arthropods.

Rab family of small GTPases: an updated view on their regulation and functions

Yuta Homma et al.

Summary: The Rab family of small GTPases plays a crucial role in regulating intracellular membrane trafficking by cycling between active and inactive states, controlled by GEFs and GAPs. Identification of new GEFs, GAPs, effectors, and post-translational modifications of Rabs have expanded our understanding of their functions beyond simple effector recruitment. Advances in CRISPR/Cas technology have accelerated knockout analyses, revealing previously unknown physiological roles of many Rabs.

Variability of an Early Developmental Cell Population Underlies Stochastic Laterality Defects

Roberto Moreno-Ayala et al.

Summary: Quantification of embryo-to-embryo variability in vertebrate development was done by studying cell number variation in the zebrafish endoderm. It was found that a sub-population of the endoderm, the dorsal forerunner cells, exhibited significantly more embryo-to-embryo variation and was associated with an increased frequency of left-right laterality defects. These fluctuations had a large stochastic component among fish of the same genetic background, suggesting a strong macroscopic phenotype resulting from stochastic variation in early development.

Rab34 GTPase mediates ciliary membrane formation in the intracellular ciliogenesis pathway

Anil Kumar Ganga et al.

Summary: Primary cilia serve as essential organizing centers for signal transduction, and defects in cilia formation can cause congenital disorders. There are two pathways for primary cilia formation – an extracellular pathway and an intracellular pathway, each with unique functional properties. Rab34 is identified as a selective mediator of intracellular ciliogenesis and plays a crucial role in ciliary membrane biogenesis.

The recycling endosome protein Rab25 coordinates collective cell movements in the zebrafish surface epithelium

Patrick Morley Willoughby et al.

Summary: In developing epithelial tissues, Rab GTPase Rab25 plays a key role in coordinating cytokinetic bridge abscission and cortical actin density, impacting cell shape changes and tissue-scale forces. Dysfunction of Rab25 leads to defective cytokinesis, resulting in anisotropic cell morphologies and delayed tissue development.

Knock-in tagging in zebrafish facilitated by insertion into non-coding regions

Daniel S. Levic et al.

Summary: A simple approach for tagging proteins in zebrafish on their N or C termini with fluorescent proteins by inserting PCR-generated donor amplicons into non-coding regions of the corresponding genes has been developed, enabling the generation of endogenously tagged alleles for crucial genes in epithelial biology and organ development. This approach facilitates the generation of knock-in lines in zebrafish, paving the way for accurate quantitative imaging studies.

Who's in control? Principles of Rab GTPase activation in endolysosomal membrane trafficking and beyond

Ann-Christin Borchers et al.

Summary: The article discusses the phenomenon of Rab cascade in the eukaryotic endomembrane system, focusing on the example of the endolysosomal system. It emphasizes the recruitment and activation of Mon1-Ccz1 in the endolysosomal and autophagic pathways, compares the principles of different GTPase cascades, and highlights the importance of regulation in organelle biogenesis and maturation.

Anisotropic expansion of hepatocyte lumina enforced by apical bulkheads

Lenka Belicova et al.

Summary: The morphogenesis of bile canaliculi in the liver is determined by the interplay between molecular pathways and mechanical forces, with hepatocytes forming specific extensions of the apical membrane that ensure anisotropic expansion of the lumen. Silencing the Rab35 gene leads to loss of apical bulkheads and cyst formation, indicating the essential role of these structures in liver tissue morphogenesis. The reengineering of hepatocyte polarity to convert bile canaliculi into epithelial tubes suggests that apical bulkheads are intrinsic mechanical elements determining the elongation of bile canaliculi during liver tissue development.

Protocol Imaging the early zebrafish embryo centrosomes following injection of small-molecule inhibitors to understand spindle formation

Abrar A. Aljiboury et al.

Summary: This protocol outlines techniques for monitoring spindle dynamics during early embry-onic divisions in zebrafish embryos, including injecting small-molecule inhibitors towards polo-like kinases, preparing embryos for three-dimensional imaging, and using confocal microscopy. These techniques are aimed at understanding how the centrosome in early zebrafish embryos constructs the mitotic spindle.
