Structural determinants of cholesterol recognition in helical integral membrane proteins

Brennica Marlow et al.

Summary: Cholesterol plays a crucial role in modulating membrane fluidity and protein function, but understanding its impact on proteins is hindered by limited and conflicting structural data. The search for cholesterol-protein interaction sites relies mainly on sequence-based algorithms, yet the necessity or sufficiency of these sites for cholesterol binding remains inconclusive. Leveraging experimental structures, this study systematically analyzes potential interaction sites and categorizes them as specific or nonspecific, aiming to enable a structure-based approach for predicting and designing cholesterol-protein interaction sites.

The Role of Cholesterol in Amyloidogenic Substrate Binding to the γ-Secretase Complex

Urszula Orzel et al.

Summary: The study showed that at high cholesterol levels, Aβ(45) was more flexible in the binding site of presenilin than Aβ(43), but the active site of presenilin at the C-terminal part of Aβ(45) was more compact, which could promote processing of this substrate. Cholesterol binding sites at low and high concentrations were also mapped, independent of typical cholesterol binding motifs.
