Quantitative Water Permeability Mapping of Blood-Brain-Barrier Dysfunction in Aging

Summary: Blood-brain-barrier (BBB) dysfunction is a key feature of aging and aging-related disorders. A study found that BBB water exchange rate (k(W)) differs in various brain regions, with higher values in the cerebral cortex compared to cerebral white matter. This difference is also observed in cerebral blood flow (CBF). Age was found to be statistically significant in relation to k(W) and CBF. Furthermore, there were moderate negative correlations between k(W) and white matter hyperintensity (WMH) volume. This research provides insights into the relationship between BBB water exchange and aging, and has implications for potential BBB-targeted therapies for aging-related brain disorders.

NVU dysregulation may lead to the entry of pathogens from the blood into the cerebral parenchyma by altering the blood-brain barrier (BBB), resulting in VCI. Additionally, the coupling of neural activity with cerebral blood flow and the clearance of metabolic byproducts may also be affected by NVU dysregulation.

Quantitative susceptibility mapping as an imaging biomarker for Alzheimer's disease: The expectations and limitations

Associations of quantitative susceptibility mapping with Alzheimer's disease clinical and imaging markers

Summary: This study investigates the associations between QSM and established markers of Alzheimer's disease. It found that increased susceptibility in deep and inferior gray nuclei is related to aging, cognitive impairment, amyloid deposition, and off-target binding of the tau ligand. Associations were also seen between susceptibility and amyloid PET, tau PET, and cortical gray matter volume in the medial temporal lobe. Additionally, iron content was found to be insufficient in the cortex for reliable detection using QSM.
