Design of enzyme-metal hybrid catalysts for organic synthesis

Summary: Enzyme-metal hybrid catalysts (EMHCs) have combined the advantages of both enzymes and metal catalysts, showing high selectivity, efficiency, and broad substrate spectrum, which are crucial for organic synthesis. Recent developments in the design and application of EMHCs have made significant progress in driving reactions that cannot be easily catalyzed by native enzymes or metals. This review summarizes the recent advances in the application of EMHCs, including enzymatic reactions, selective chemical transformations, and chemoenzymatic cascade reactions catalyzed by biohybrid catalysts, and discusses the structure-function relationships of EMHCs. Lastly, the perspectives and limitations in this field are highlighted, and research directions for this exciting theme are proposed.

Arylative Allenol Cyclization via Sequential One-pot Enzyme & Palladium Catalysis

Summary: The one-pot combination of halogenation biocatalysis and Suzuki-type cross coupling enables the direct arylative cyclization of allenic alcohols with boronic acids. A novel heterogeneous nanobiohybrid was developed, consisting of an oxidase matrix hosting small spherical palladium nanoparticles, exhibiting catalytic competence for both the biocyclization as well as the C-C bond-forming cross coupling, underlining the potential of this new techniques for streamlining chemoenzymatic approaches.

Clicked Bifunctional Dendrimeric and Cyclopeptidic Addressable Redox Scaffolds for the Functionalization of Carbon Nanotubes with Redox Molecules and Enzymes

Summary: Carbon nanotube electrodes were modified with ferrocene and laccase using click reactions strategies involving diazonium functionalization and oxime ligation, achieving a high surface coverage of ferrocene. The immobilization of active multicopper enzymes, laccases, on surfaces via copper(I)-catalyzed azide-alcyne cycloaddition was also successful, highlighting the efficiency of oxime-ligated dendrimers or cyclopeptides. Despite controlled orientation, this immobilization strategy was found to impact electron tunneling with enzyme active sites.

Electron Transfer to the Trinuclear Copper Cluster in Electrocatalysis by the Multicopper Oxidases

Summary: High-potential multicopper oxidases (MCOs) are efficient catalysts for oxygen reduction with low overpotentials and commonly used in biofuel cells. This study investigated the electron transfer pathways in MCO electrocatalysis and found that electron transfer primarily occurs through the T1 site to the trinuclear copper cluster (TNC). The research provides insights into the factors controlling the kinetics of MCO electrocatalysis and guidance for designing more efficient biocathodes for the ORR.

Incorporation of Metals and Enzymes with Porous Imine Molecule Cages for Highly Efficient Semiheterogeneous Chemoenzymatic Catalysis

Hybrid chemoenzymatic heterogeneous catalyst prepared in one step from zeolite nanocrystals and enzyme-polyelectrolyte complexes

Summary: The development of hybrid chemoenzymatic heterogeneous catalysts combining inorganic catalysts and enzymes is a promising strategy to improve the efficiency of chemoenzymatic reactions. A novel nanostructured hybrid catalyst consisting of glucose oxidase and TS-1 zeolite was successfully prepared in one step using continuous spray drying, enabling biocatalytic formation of H2O2 and subsequent epoxidation of allylic alcohol. The use of enzyme-polyelectrolyte complexes (EPCs) increased enzyme stability, prevented enzyme leaching, and stabilized the enzyme against pH changes, leading to unprecedented performance in the targeted chemoenzymatic reaction.
